摘 要:目的探讨脑创伤(TBI)大鼠钾-氯协同转运蛋白2(KCC2)和γ-氨基丁酸(GABA)的表达变化及亚低温治疗对其影响。方法将30只Wistar大鼠随机分为对照组(C组)、创伤组(TBI组,给予液压冲击,不做处理)和治疗组(T组,液压冲击后接受亚低温治疗,32℃),亚低温干预在创伤后即刻给予32℃低温处理持续6 h。高效液相色谱法测定GABA水平,酶联免疫吸附试验检测各组大鼠血清KCC2水平;反转录-聚合酶链反应、Western blotting检测各组大鼠脑组织中KCC2 mRNA和蛋白的表达。结果与C组比较,T组、TBI组大鼠血清KCC2水平、脑组织KCC2蛋白及mRNA表达均显著降低(P<0.05);T组大鼠血清KCC2水平较TBI组显著增高,差异有统计学意义(P<0.05);GABA变化趋势与KCC2相反。结论 KCC2在大鼠脑创伤过程中表达降低,亚低温进行干预可上调KCC2的表达、下调GABA表达。Objective To investigate the changes of K^+-Cl^-cotransporters-2( KCC2) and gamma-aminobutyric acid(GABA) and the protecting effect of mild-hypothermia after traumatic brain injury(TBI)in rat model. Methods Adult Wistar rats were randomly divided into control group(group C),traumatic brain injury group(group TBI,accepted sap pressure impact and no treatment) and treatment group(group T,accepted mild-hypothermia treatment after sap pressure impact,32 ℃). Mildhypothermia intervention for 6 h immediately initiated after TBI in group T. GABA was determined by high performance liquid chromatog-raphy method. The changes of KCC2 in serum were determinated by enzyme linked immunosorbent assay. KCC2 mRNA and protein expression in brain tissue were examined by reverse transcription-polymerase chain reaction and Western blotting,respectively. Results The serum level of KCC2,KCC2 mRNA and protein expression in brain tissue decreased significantly in group TBI and group T compared with group C(P〈 0. 05),which increased significantly in group T compared with those in group TBI( P〈0. 05). The trend of GABA was reversed. Conclusion Mild-hypothermia treatment,which plays a neuroprotective role,might decrease the level of GABA and increase the level of KCC2 induced by TBI in rats.
