免疫不孕奶牛与健康奶牛宫颈黏液蛋白表达及初步分析

摘  要:为研究不孕与可孕奶牛间宫颈黏液蛋白质的差异,揭示奶牛宫颈黏液蛋白质与不孕的关系,采用二维凝胶电泳技术获得了不孕与可孕奶牛宫颈黏液中总蛋白的表达图谱,经过Ymension Revolutionary 2DGE Software自动匹配分析,得出不孕奶牛宫颈黏液蛋白质上样量分别为0.8 mg和1.6 mg时,蛋白质斑点数分别为134个和151个,检测出具有明显表达差异蛋白斑点数为17个。不孕与可孕奶牛宫颈黏液蛋白质上样量为1.6 mg时,蛋白质斑点数分别为151个和67个,有62个匹配的蛋白质斑点。证明不孕奶牛与可孕奶牛宫颈黏液蛋白中存在差异表达的蛋白斑点。该试验将有助于解释免疫性不孕奶牛宫颈黏液中的差异蛋白对生殖的影响,为改善诊断的特异性、治疗免疫性不育提供理论基础。To further explore the relationship between dairy cows cervix mucus and the infecundity at protein level and analyze the expression changes of proteins in cervix mucus of fertile and infertile dairy cows,the proteome expression profile of the fertile and infertile dairy cows' cervix mucus were obtained by 2D-PAGE.When the cervix mucus sample quantity were 0.8 mg and 1.6 mg,the number of the protein spot were 134 and 151 respectively,the differential protein expression was 17.When the fertile and infertile dairy cows' cervix mucus sample quantity were 1.6 mg,the number of the protein spot were 151 and 67 respectively,the number of the match protein spot was 62.The results indicated that there were significant differences in expression protein between fertile and infertile dairy cows,and valuable information might be provided to find new regulation markers of mastitis and potential protein targets for treatment.
