摘  要:目的:用高通量Arraystar T-UCR芯片研究超保守区非编码RNA(T-UCR)在银屑病外周血单个核细胞(PBMC)中的差异表达谱。方法:抽取7例寻常型银屑病患者及3例正常人的外周血,分离PBMC后提取总RNA,将总RNA纯化,转录合成荧光标记的cRNA,cRNA样本片段化后与芯片杂交,使用Agilent G2505C芯片扫描仪扫描芯片,配合数据处理软件读取、处理数据,筛选出1.5倍及以上差异表达基因。结果:共有109种已知T-UCR在PBMC中表达,银屑病组与健康对照组相比,有5个T-UCR呈共同差异表达,其中上调1个,下调4个;有2 043种可能是新的T-UCR在PBMC中表达,共182种呈共同差异表达,其中上调100个,下调82个;与UCR区域重叠的mRNAs和UCR邻近基因分别有232个和67个表达有差异,Gene Ontology分析发现在参与细胞生理过程的36个差异表达的与UCR区域重叠的mRNAs中,有5个参与T细胞免疫活性相关的生物过程,例如T细胞活化、免疫反应中T细胞的分化等。结论:T-UCR可能参与寻常型银屑病的发病。Objective:To investigate differential expression profile of transcribed ultra con-served region ( T-UCR) in PBMC of patients with psoriasis vulgaris by microarray analysis .Meth-ods:Blood samples for extraction of total RNA in PBMC were collected from 7 patients with psoria-sis vulgaris and 3 healthy individuals.mRNAs and LncRNAs were purified from total RNA.Then, each sample was amplified and transcribed into fluorescent cRNA .The labeled cRNAs were hy-bridized onto the human T-UCR Array.After the slides were washed , the arrays were scanned by the Agilent Scanner G2505C.Results:A total of 109 T-UCRs were found expressed in PBMC . Compared with T-UCR in healthy individuals , 1 was upregulated and 4 T-UCRs were down-regula-ted in patients with psoriasis vulgaris .A total of 2 043 novel T-UCRs were found expressed in PB-MC.Among them, 100 was upregulated and 82 were down-regulated in patients with psoriasis vul-garis.Both of 232 RNA transcripts whose transcription Units overlap UCRs and 67 UCR-proximal genes were significantly differentially expressed .GO analysis revealed that 36 differentially ex-pressed RNA transcripts whose transcription Units overlap UCRs were enriched in cellular process , and 5 of them were related to T cells .Conclusion:T-UCR may play an important role in the path-ogenesis of psoriasis vulgaris .
