摘  要:目的探讨内源性salusin-β在脓毒症大鼠脾、胃、小肠、下丘脑组织及血清中的分布及含量变化。方法雄性SD大鼠36只,采用单纯随机抽样的方法随机分为假手术组(n=9)和脓毒症模型组(简称"模型组",n=27)。假手术组仅翻动盲肠,模型组以盲肠结扎穿刺术(CLP)制作脓毒症模型,于造模后6 h、12 h及24 h时分别处死9只大鼠,取脾、胃、小肠及下丘脑,并留取血清标本。采用酶联免疫吸附试验测定salusin-β含量。结果 1假手术组大鼠脾、胃、小肠、下丘脑及血清中均含有一定量salusin-β,且脾组织中的含量最高,明显高于其他组织(P<0.05)。2与假手术组比较,模型组CLP后6 h时大鼠脾、小肠、胃及血清中salusin-β水平开始升高且有统计学意义(P<0.05),其中脾组织和血清中在12 h时达最高,小肠组织中在24 h时达最高;胃组织中无明显变化;下丘脑组织中salusin-β水平虽在术后6 h时开始升高,仅在12 h时有统计学意义(P<0.05)。结论脓毒症大鼠体内salusin-β含量随时间依赖性改变,提示salusin-β可能与脓毒症有一定的关系。Objective To investigate the distribution and content of endogenous salusin-β in septic rats. Methods Thirty-six SPF male SD rats were randomly divided into sham operation group (n=9) and septic model group (n=27). Only the cecum was turn in the sham operation group and the septic model was made by the cecal ligation and puncture (CLP) in the septic model group. The rats were sacrificed at 6 h, 12 h, and 24 h after modeling in the septic model group. The contents of salusin-β in the tissues of spleen, stomach, small intestine, hypothalamus, and serum specimens were detected by enzyme-linked immunosorbent assay. Results ① The salusin-β endogenously generated in the rat tissues including the spleen, stomach, small intestine, hypothalamus, and serum. The content of salusin-β in the spleen tissue was higher than that in the other tissues (P〈0. 05). ② The contents of salusin-β in the spleen, stomach, small intestine tissues together with the serum increased significantly at 6 h after CLP as compared with the sham operation group (P〈 0.05). The contents of salusin-β in the spleen tissue and serum were peaked at 12 h, in the small intestine tissue reached the summit at 24 h. While, the content of salusin-β had no significant fluctuation in the stomach tissue. The content of salusin-β began to increase at 6 h in the hypothalamus tissue, and significantly increased at 12 h after CLP (P〈0. 05). Conclusion The time-dependent change of salusin-β in sepsis rats suggests that salusin-β might be involved with the pathogenesis of sepsis.
