摘  要:目的观察通心络对糖尿病小猪心肌组织中转化生长因子β1(TGF-β1)表达的影响,探讨糖尿病心肌病的发病机制及通心络的保护作用。方法制备糖尿病小型猪模型,随机分为糖尿病心肌病组(DCM)和通心络治疗组(TXL),以正常小猪作为对照组(C)。检测各组小猪的血糖变化情况。观察通心络对糖尿病小猪血流动力学的影响。应用免疫组化技术和Western blot技术观察各组动物心肌组织中TGF-β1的表达。结果通心络对糖尿病小猪的血糖无明显影响,但是可明显增加糖尿病小猪心肌血流。与对照组相比,糖尿病组小猪心肌组织中TGF-β1的表达明显增加,通心络可显著降低TGF-β1的表达。结论通心络可改善糖尿病心肌组织的功能,可能通过降低TGF-β1表达这一途径改善心肌纤维化状态,对糖尿病心肌组织具有保护作用。Objective To observe the effect of a Chinese medicine preparation, Tongxinluo, on the expression of TGF - β1 in diabetic porcine myocardium, and to investigate the pathogenesis of diabetic cardiomyopathy and effect of Tongxinluo on this disease. Methods Nine 5 - 6 months old Bama minipigs were used in this study. Diabetic minipig models were generated by intravenous injection of streptozotocin and alloxan. The diabetic pigs were randomly divided into diabetic cardiomyopathy (DCM) and Tongxin- luo (TXL) treatment groups ( n = 3 ). Three normal pigs were used as control group ( C ). Blood glucose was detected. The effect of tongxinluo on hemodynamica of diabetic pigs was observed. Immunohistochemitry and Western blot were used to observe the expression of TGF - β1 in myocardial tissue. Results Tongxinluo had no significant effect on the blood glucose of diabetic pigs, but increased the blood flow in myocardi- urn. Compared with the control group, the expression of TGF -β1 in the myocardium of diabetic pigs was significantly increased, while Tongxinluo significantly reduced the expression of TGF - β1 in their myocar- uo may improve the function of diabetic myocardium. It may be improve the state of myocardial fibrosis by reducing the expression of TGF - β1 and has a protective effect on the diabetic myocardium.
