摘 要:目的探讨S-HPM辐射致X、Y精子的损伤及其差异。方法经流式分选得到高纯度的牛X、Y精子,经30 mW/cm2和100 mW/cm2的S-HPM辐射4 min,通过精子动态分析系统检测辐射后1 h、2 h、4 h、6 h、12 h和24 h的X精子和Y精子的活动度的差别;吖啶橙/溴化乙啶(acridine orange/ethidium bromide,AO/EB)染色检测辐射后1 h、6 h、12 h和24 h的X精子和Y精子凋亡率的变化。透射电镜观察100 mW/cm2S-HPM辐射后4 h X、Y精子超微结构损伤程度的差异。顶体酸性磷酸酶染色检测辐射后1 h、6 h和12 h顶体功能的变化差异。结果经30 mW/cm2和100 mW/cm2的S-HPM辐射后,精子活动度均随时间延长呈降低趋势,X、Y精子活动度的降低未见显著性差别(P>0.05)。S-HPM辐射可致X、Y精子凋亡率增加,呈现Y精子重于X精子,100 mW/cm2重于30 mW/cm2的趋势,其中早期具有显著性差异(P<0.05)。超微结构显示S-HPM辐射后精子以头部病变为主,尾部病变不明显,Y精子损伤重于X精子。S-HPM辐射后精子顶体酸性磷酸酶含量降低,Y精子重于X精子,100 mW/cm2重于30 mW/cm2(P<0.05,P<0.01)。结论 S-HPM辐射后X、Y精子均发生损伤,以精子头部病变为主,精子顶体酸性磷酸酶含量降低,Y精子的损伤重于X精子。Objective To explore the damaging differences of S-HPM (S band high power microwave radiation) on X and Y sperms. Methods X and Y sperms were isolated from mature cow, and ex- posed to 30 mW/cm^2 and 100 mW/cm^2 S-HPM for 4 min. SCA sperm dynamic analysis systerm was used to decteet the sperm motility. AO/EB staining was used to determine germ cells apoptosis. Trans- mission electron microscope (TEM) was used to observe the ultrastructural changes. APC staining was used to examine the ACPase content of apical body. Results 2 - 24 h after 30 mW/cm^2 and 100 mW/cm^2 S-HPM radation,X and Y sperms mobility showed a reduetive trend with the time lasting, but no significant differences were found on irradiated X and Y sperms. Apoptotic and necrotic rates of X and Y sperms showed an incremental trend, and rates of apoptosis and necrosis in 100 mW/cm^2 radia- tion group were obvious than those of 30 mW/cm^2 radiation group, and at the same time these incremen- tal trend in Y group was more clearly than that of X group. Uhrastructural changes showed that concen- trate and inhomogeneous nucleus were seen in the head of X and Y sperms, vacant spaces between the endomembrane and outer membrane were enlarged, and the cytoplasmic membranes were circuitied and disrupted. These processes were severer in Y sperms than that of X sperms. No apparente changes and differences were found in each end part of X and Y sperms. After S-HPM radiation, decreases of acid phosphatase content in acrosome were more obvious in Y sperms and in 100 mW/cm^2 group than those in X sperms and in 30 mW/cm^2 group(P 〈 0.05 or P 〈 0.01 ). Conclusions After S-HPM radiation, injures are observed both in X sperms and Y sperms, and major damages are found in their heads, and reduction of acid phosphatase content in acrosome is more evident in Y sperms than in X sperms, which show relationships of time effect and dose effect
