摘 要:目的:研究纳米氧化铈对大鼠肺泡巨噬细胞NR8383的毒性作用和对细胞的增殖影响;考察纳米氧化铈作用于细胞的生物效应是否存在剂量和粒径效应。方法:不同终浓度200、100、50、20、10、5、2.5、1.25、0.625μg/m L的20.8 nm Ce O2-NPs体外作用于96孔板中NR8383细胞24 h,采用CCK-8试剂盒检测细胞活力;四种粒径20nm、50-100 nm、300-500 nm、1-5μm Ce O2分别以四种终浓度100、50、20、10μg/m L作用于NR8383细胞24 h,采用CCK-8试剂盒检测检测细胞活力。结果:不同浓度20.8 nm Ce O2-NPs作用于NR8383细胞,与对照组相比,除1.25、2.5、5μg/m L外,其他浓度下细胞存活率随浓度增大而升高,100、200μg/m L最明显;有剂量效应。四种粒径四种浓度的纳米氧化铈作用于NR8383细胞,50-100 nm的20μg/m L浓度和1-5μm的50、10μg/m L浓度Ce O2-NPs抑制细胞增殖,但不明显;其他各组均促进细胞增殖,其中20 nm的20μg/m L,50-100 nm的50μg/m L、100μg/m L最明显。剂量尺寸效应不明显。结论:小粒径纳米氧化铈对NR8383细胞增殖有促进作用,无明显毒性;微米级氧化铈有些浓度下细胞存活率降低,可能对细胞造成毒性。Objective:To investigate the toxicity effect of nano cerium oxide on alveolar macrophages NR8383 and its effect on the proliferation of NR8383 as well in rats and to verify whether there is a effect relation between the dose and particle size in the biological effect that nano cerium oxide bring about on the cells. Methods:20. 8 nm cerium oxide - NPs of different final concentrations(200,100,50,20,10,5,2. 5,1. 25 and 0. 625 μg/ mL)were applied respectively to 96 - well plates in vitro where NR8383 cells were cultivated for 24 h,with the cell activity tested by CCK - 8 kit. CeO2 of four different particle sizes (20 nm,50 - 100 nm,300 - 500 nm and 1 - 5 μm)were applied to NR8383(24 h)at four different final concentrations(100, 50,20,10 μg/ mL),with the cell activity tested by CCK - 8 kit. Results:Compared with that in the control group,the cell survival rate increased along with the rise in the concentration of CeO2 - NPs at different concentrations except at the concentra-tions of 1. 25,2. 5 and 5 μg / mL when 20. 8 nm CeO2 - NPs of different concentrations worked on NR8383 cells,most signifi-cantly at the concentrations of 100 and 200 μg/ mL,with a dose effect. Except for 50 - 100 nm CeO2 - NPs of 20 μg / mL and 1 - 5 μm CeO2 - NPs of 50 and 10 μg/ mL,which could insignificantly inhibited cell proliferation,CeO2 - NPs of other particle sizes and other concentrations all could promote cell proliferation while working on NR8383 cells,with the effect of 20 nm at 20 μg / mL and 50 - 100 nm at 50 μg/ mL and 100 μg/ mL most obvious and with dose - size effect not obvious. Conclusion:Nano cerium oxide of small sizes has an active effect on the proliferation of NR8383 cells,with no significant toxicity. Micron grade cerium oxide,at certain concentrations,induces lower cell viability and may cause toxicity to cells.
