摘 要:目的检测创伤后应激障碍(PTSD)大鼠杏仁核内质网分子伴侣钙网蛋白(CRT)表达的变化。方法建立无连续单一刺激(SPS)模型,随机将大鼠分为SPS刺激后1 d组(SPS1d)、7 d组(SPS7d)、14 d组(SPS14d)及正常对照组。应用免疫组织化学、逆转录聚合酶链反应及透射电镜技术,检测各组杏仁核神经元CRT表达变化及观察神经元超微结构的改变。结果与正常对照组比较,SPS各组神经元CRT阳性表达均增加(P<0.001),尤以SPS7d组最为明显。SPS各组神经元细胞核及粗面内质网均发生不同程度的改变。结论 SPS可诱导PTSD样大鼠过度内质网应激,引发杏仁核CRT表达上调并通过内质网径路发生细胞凋亡。Objective To observe the changes of Calreticulin (CRT) in the amygdaloid nucleus of rats.Methods The rats were randomly divided into four groups,one group served as normal control,while others were induced for single-prolonged stress (SPS)model as SPS groups (1 d,7 d,14 d).Immunohistochemistry and RT-PCR were formed to examine the changes of CRT in the amygdaloid nucleus of rat ; the neuron ultrastructure of amygdaloid nucleus was detected by transmission electron microscope.Results The expression of CRT in the amygdaloid nucleus increased after SPS stimulation comparing with the normal control group (P < 0.001),and reached the peak in the group of SPS 7 d.There were obvious changes of neuron nucleus and the rough endoplasmic reticulum of amygdaloid nucleus at SPS groups.Conclusion PTSD induced endoplasmic reticulum stress in the amygdaloid nucleus and enhanced the expression of CRT after SPS stimulation.SPS stimulation also induced apoptosis through endoplasmic reticulum pathway in the amygdaloid nucleus.
