摘 要:目的:建立大豆黄卷总黄酮的测定方法,同时对其水煎煮过程中的提取动力学过程进行模拟,建立动力学方程并验证。方法:采用紫外分光光度法,筛选对照品和测定波长,并对其方法学进行考察;使用该方法,测定不同提取时间和溶剂下大豆黄卷水提浓度,以简化的数学动力学模型为基础,推算出方程参数,将测定结果代入进行方程拟合。结果:染料木苷在0.0009296~0.01162 g·L-1具有良好的线性关系,回归方程为Y=54.859X-0.0034,r=0.9999,平均回收率为103.6%,RSD=0.83%;拟合的动力学模型结果为CB=(0.1520×t0.5/M-1.7970)0.5807,验证结果良好。结论:所建立的大豆黄卷总黄酮测定方法稳定可行,重现性好。模型适用并能良好的描述大豆黄卷水提取的动力学过程。Objective: To establish the determination method of total flavonoids in Geminated Soybean and study its extraction process of decocting,deduce the kinetic model and test its accuracy. Methods: Ultraviolet Spectrophotometry was used to choose the reference substance and the detection wavelength. And the method was used to determine concentration of total flavonoids in different extraction time( t) and solvent loads( M). Based on the simplified model,correlation parameters were deduced and the equation was set up. Results: Genistin had good linear relation in the range of 0. 0009296-0. 01162 g L- 1and the average recovery was 103. 6% and RSD was 0. 83%. Its kinetic model was: CB=( 0. 1520 × t^0. 5/M- 1. 797 0)0. 5807 and the acurancy was tested. Conclusion: The method is simple,quick,stabile and reproducible and the kinetic model can describe the extraction process of Geminated Soybean.
