摘  要:目的:研究扶正散结汤拆方配伍中药对Lewis肺癌荷瘤小鼠肿瘤转移及血管生成机制影响的差异及其协同作用。方法:80只C57小鼠,随机均分为空白组、模型组、顺铂组、扶正组、化痰组、活血组、解毒组、联合组,除空白组外其余小鼠右腋皮下接种Lewis瘤细胞建立肺癌模型,从接种后的第2天开始分别给予相应药物灌胃或腹腔注射,连续14 d后处死,将瘤体剥离常规固定、脱水、包埋切片,免疫组化SP法检测肿瘤组织中VEGF、ES、MMP-2。结果:各给药组肺转移结节数量均低于模型组(P<0.05),以顺铂组和联用组为最少、也最接近,其次依次为扶正组、解毒组和活血组,各组与顺铂组相比,除化痰组外,无显著性差异。顺铂及各配伍组分组均较模型组VEGF、MMP-2表达量降低、ES表达量升高,其中解毒组VEGF表达最低(P<0.05)、联用组ES表达量最高(P<0.05 or P<0.01)、MMP-2表达量最低(P<0.05),而顺铂组不具任何优势。结论:扶正散结汤拆方配伍四法联用具有更好抗肿瘤转移效果,与顺铂相当;在抗肿瘤微血管生成机制方面,各配伍组分作用各有特点,但四法联用具有更优的效果。Objective: To study the Fuzheng Sanjie Recipe and its separated prescriptions on the lung cancer metastasis and angiogenesis differences and synergetic action of Lewis lung carcinoma. Method: Eighty SPF C57 BL /6 mice were randomly and equally divided into 8 groups: normal group,model group,DDP group,Yiqi Fuzheng group,Huatan group,Huoxue group,Qingjie group,Combination group. We established ubcutaneous cancer models in C57 BL /6 male mice by inoculation of Lewis lung carcinoma except normal group. Each group was given intragastrical administration and once a day continually for 14 days after inoculation. A portion of the tumors by 10% neutral formalin- fixed and paraffin- embedded after dehydration were for the tumors Immunohistochemical( IHC) testing. Result: The number of tumor lesions on lung metastasis in treatment group had significant difference comparing with model group( P〈0. 05). The DDP group and combination group were the least and closest to each other of the number of tumor lesions,followed by Fuzheng group,Qingjie group and Huoxue group. No significant changes were demonstrated with treatment group except Huatan group.Compared with model group,the expression of DDP group and the separating prescriptions groups 'quantity were decreased significantly in VEGF,MMP- 2 and increased in ES( P〈0. 05). The VEGF of Jiedu group and the MMP- 2 of the combination group reached the minimum and the ES of the combination group reached the maximum among these groups( P〈0. 05 or P〈0. 01). The DDP group did not have advantages but other groups had. Conclusion: The combination group of Fuzheng Sanjie recipe's separating prescriptions was effective as the DDP group with. Each separating prescription had a complementary inhibitory action on tumor angiogenesis but the combination group was more effective.
