摘 要:目的:观察虚寒证大鼠体质量、体温、自主活动及皮肤血流量变化,探讨热药吴茱萸对虚寒证大鼠的干预作用。方法:大鼠先大剂量后小剂量皮下注射利血平造模,造模第13天始分别按10mL/kg灌胃吴茱萸水煎液0.42、0.21g生药/mL,模型对照组与空白对照组给予等容量0.9%氯化钠溶液,连续12d;测定各组动物造模前、造模第1、3、6、9、12天及给药第1、4、7、9、12天体质量、体温;测定各组大鼠造模第12天、给药第12天自主活动次数及耳部微循环血流量。结果:模型对照组大鼠体质量减轻,体温下降,自主活动次数及耳部微循环血流量减少;吴茱萸高、低剂量组可明显对抗虚寒证大鼠体质量减轻和体温降低,可明显增加其自主活动次数及耳部微循环血流量。结论:热性中药吴茱萸可较好地改善虚寒证大鼠症状,该模型可用于中药寒热药性的研究。Objective: To observe the body weight, body temperature, the skin microcirculation blood flow of rat ear and the autonomic activities of asthenia cold syndrome rats induced by reserpine. To discuss the effects of Fructus Evodiae on asthenia cold syndrome rat model. Methods: Male SD rats were randomly divided into a blank control group, a model control group, Fructus Evodiae high and low dose group. The model control group and the drug treatment groups accepted hypodermic injection with reserpine on the back, at the dose of 0.5mg/kg at the first 1-8 days and 0.3mg/kg at 9-12 days, once a day for 12 days. After model established, the drug treatment groups were intragastric administration with the water decoction of Fructus Evodiae crude drug at the dose of 0.42, 0.21g/mL, 10rnL/kg, once a day for 12 days, while the model control group and blank control group with the same amount of normal saline. The body weight and the rectal temperature were measured at 0, 1, 3, 6, 9, 12d, as well at 1, 4, 7, 9, 12d after model established. The skin microcirculation blood flow of the rat ear and autonomic activities were measured at the time point of 12d and at 12d after model established. Results: The body weight, the rectal temperature, the skin microcirculation blood flow of rat ear, and the autonomic activities reduced in model control group, while the body weight, the rectal temperature, the skin microcirculation blood flow of rat ear, and the autonomic activities significantly increased in Fructus Evodiae high and low dose group. Conclusion: Fructus Evodiae has good effects on the asthenia cold syndrome rat model. The asthenia cold syndrome rat model induced by reserpine could be used in study of cold-heat nature of Chinese herbal medicine.
