摘  要:目的:考察黄芪部分有效部位(黄酮、多糖、皂苷)对糖尿病大鼠血糖、超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的影响,阐释黄芪有效部位之间的交互关系。方法:将126只大鼠按血糖随机分为9组,每组14只,除正常组外,其余各组用链脲佐菌素造模后按2×2×2析因设计实验方案实施实验。测定血糖及血清SOD、MDA、TNF-α、IL-6等指标,有关联的指标计算综合评分;采用单因素方差分析和析因设计(重复测量资料)方差分析等方法,分析黄芪3个有效部位的单独效应及交互效应。结果:模型组与正常组比较,各项指标差异均有统计学意义(P<0.05)。黄酮有显著降血糖作用(P<0.05),多糖、皂苷以及药物间的交互作用均不显著;黄酮对SOD、MDA作用显著(P<0.05),黄酮与皂苷、多糖与皂苷合用效果均优于单一用药。黄酮、多糖、皂苷对TNF-α,IL-6均有影响,黄酮与多糖合用优于单一用药,黄酮与皂苷合用不如单用黄酮。结论:黄芪黄酮有降血糖作用,与多糖、皂苷合用效果不明显;黄芪黄酮抗氧化作用明显,黄酮与皂苷、多糖与皂苷合用均能提升单一用药效果;在降低TNF-α、IL-6方面,黄酮与多糖合用效果最佳。Objective: To investigate the effects of main effective parts of Astragalus (total flavonoids, polysaccharides, saponins) on blood glucose, superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor alpha (TNF-α), interleukin-6 (IL-6) in diabetic rats, and to explain the interrelationship between the effective parts of Astragalus. Methods: 126 rats were randomly divided into 9 groups, with 14 rats in each group. Except the normal group, rest groups induced by streptozotocin to establish the model, 2×2×2 factorial design experiment scheme implemented in this experiment. The blood glucose, SOD, MDA, TNF- α, IL-6 were detected, the correlation index of comprehensive score was calculated. Three effective parts of Astragalus and their interaction were analyzed by using single factor analysis of variance and factorial design (repeated measurement data) variance analysis methods. Results: Compared with the normal group, the indexes had significant differences in the model group (P〈0.05). Flavonoids had a hypoglycemic effect (P〈0.05). Polysaccharides, saponins and interactions between drugs were not significant. The effect of flavonoids on SOD, MDA was obvious (P〈0.05), The effect of flavonoids and saponins, polysaccharides and saponins in combination was better than a single drug. Flavonoids, polysaccharides, saponins on TNF-α, IL-6 had a significant effect, flavonoids and polysaccharides in combination was better than a single drug, flavonoids and saponins in combination was not better than flavonoids. Conclusion: The flavonoids Astragalus had hypoglycemic effect, the effect of combination medication was not obvious. The antioxidant effect of Astragalus flavonoids was significant, flavonoids and saponins, polysaccharides and saponins in combination could enhance the effect of a single drug. Flavone and polysaccharide in combination could achieve the best effect in reducing IL-6 and TNF- α.
