摘  要:目的:探讨红景天苷对β淀粉样蛋白(Aβ)所致SH-SY5Y细胞损伤的保护作用及其可能机制。方法:通过体外培养SH-SY5Y细胞,采用含Aβ1-40的细胞培养基制备疾病模型,经Aβ1-40作用24h后,不同浓度100和200μmol/L红景天苷干预,MTT法检测各组细胞存活率;流式细胞技术、TUNEL检测细胞内活性氧ROS及凋亡情况;ELISA检测细胞内AGEs含量;Western blot检测细胞内晚期糖基化终末产物受体RAGE、NF-κB(pp65)、Bax和Bcl-2蛋白的表达变化。结果:红景天苷干预可明显改善Aβ1-40损伤后的细胞存活率,抑制Aβ1-40引起的细胞内ROS水平及细胞凋亡率升高;降低RAGE、NF-κB(p-p65)蛋白水平表达,逆转Aβ1-40导致的细胞Bcl-2/Bax蛋白表达的比例下降。结论:红景天苷可以抑制Aβ1-40诱导SH-SY5Y细胞损伤及凋亡,其机制可能与调节RAGE、NF-κB(p-p65)及相关凋亡基因Bax和Bcl-2的蛋白表达有关。Objective: To investigate the effect of salidroside on 13-amyloid (Aβ) SH-SY5Y cells induced injury and its possible mechanism. Methods: SH-SY5Y cells were cultured in vitro, using cell culture medium containing Aβ1-40, for preparing disease models, cultured with Aβ-40 for 24h, different concentrations of salidroside (100 and 2001amol/L) were used for the cure, MTT (Tetramethyl triazole salt) assay was used for checking the cell viability; Flow cytometry, TUNEL were used for the detection of intracellular reactive oxygen species ROS and apoptosis; The detection of intracellular AGEs content by ELISA; The detection of intracellular advanced glycation end products receptors RAGE, NF-κB (p-p65), Bax and Bcl-2 protein expression changes were used by Western blot. Results: Salidroside intervention can significantly improve the cell viability after injuried by Aβ1-40, inhibit the increased intracellular ROS level and apoptosis rate induced by Aβ1-40; it can reduce expression of RAGE, NF-xB (p-p65), also reverse Bcl-2/Bax protein ratio decline induced by Aβ1-40. Conclusion: Salidroside can inhibit the damage and apoptosis of SH-SY5Y ceils induced by Aβ1-40, which may be related adjustment of ROS, AGEs/RAGE/NF-r,B axis and apoptosis related genes Bax and Bcl-2 protein expression.
