摘  要:目的:观察鹿茸归经与骨关节炎软骨靶器官TGF-βRⅠ、TGF-βRⅡ的关联性。方法:选择3月龄健康雌性SD大鼠100只,体质量(200±20)g。常规喂养1周后,采用经典Hulth方法造模,造模后6周,行病理学观察,确认造模成功,按体质量分层随机数字表法分成4组:鹿茸低剂量组、鹿茸高剂量组、盐水组、模型组,于灌胃后2、4、6周,分别取大鼠双侧膝关节软骨,采用荧光定量PCR和蛋白免疫印迹杂交方法分别检测TGF-βRⅠm RNA、TGF-βRⅡm RNA和蛋白表达。结果:各组大鼠TGF-βRⅠ、TGF-RβⅡm RNA和蛋白随时间变化的趋势有显著差异(P<0.01);与模型组比较,鹿茸低、高剂量组TGF-βRⅠm RNA、TGF-βRⅡm RNA和蛋白的表达量显著增加(P<0.01)。结论:鹿茸通过归经靶向作用于软骨细胞膜上的TGF-βRⅠ、TGF-βRⅡ受体,引起软骨靶器官组织受体的表达上调,促进了TGF-β信号通路的转导而起到修复软骨的作用。软骨细胞膜上的TGF-βRⅠ和TGF-βRⅡ受体可能是鹿茸作用的靶点之一。Objective: To observe the correlation between the channel tropism of the pilose antler and TGF- β R I, TGF- βR Ⅱ of cartilage target organ in early osteoarthritis (OA). Methods: 100 healthy female SD rats of 3 months old and with the weight of (200±20)g were selected and the animal model was established follow the Hulth method after routine feeding for one week. After six weeks, the pathology technique was used to verify the animal model, which was successfully established. Then the rats were randomly divided into 4 groups: low-dose group of the pilose antler, high-dose group of the pilose antler, control group and model group. The cartilage of knee articular was taken in the second, fourth and sixth week after intragastric administration. Western blot and quantitative PCR were used to test the protein and mRNA expression of TGF- β R I and TGF- β R Ⅱ. Results: 100 were involved in the result analysis. The result of pathology technique showed that the animal model was established successfully. As the extension of the intragastric administration time, the expression of protein and mRNA of TGF- βR I and TGF- β R Ⅱ gradually changed (P〈0.01). Compared with the model group, the expression of TGF- β R I and TGF- β R Ⅱ mRNA and protein in the rats of low and high-dose group of the pilose antler increased significantly (P〈0.01). Conclusion: Pilose antler could promote the expression of TGF- β R I and TGF- β R Ⅱ receptor through channel tropism. It could repair cartilage through promoting the transmission of TGF- β signaling pathway. The TGF- β R I and TGF- β R Ⅱ receptor of cartilage membrane might be one of the targets of treating osteoarthritis with pilose antler.
