摘  要:目的:探讨参附注射液对阿霉素所致大鼠心肌细胞损伤的防治作用。方法:将大鼠随机分为正常对照组、模型组和参附注射液低、中、高剂量组(1、2和4m L·kg-1·d-1),每组14只。模型组于实验开始后第2、4天腹腔注射阿霉素1mg/kg,第6、8天腹腔注射阿霉素2mg/kg,第10、12天腹腔注射阿霉素3mg/kg,第14、16天腹腔注射阿霉素4mg/kg,累计用药剂量达20mg/kg;参附组于造模后1周,按低、中、高剂量组分别腹腔注射参附注射液1、2、4m L/kg,每日1次,共2周。采集心室肌组织,应用光镜观察心肌病理形态学变化;Annexin V/PI双染色法检测心肌细胞凋亡情况;Western blot检测心肌细胞Bcl-2、Caspase-3蛋白的表达。结果:模型组和参附注射液高剂量组心肌细胞早期凋亡率分别是(30.15±1.26)%,(12.74±1.03)%;Bcl-2蛋白表达分别是(0.40±0.04),(1.33±0.01);Caspase-3蛋白表达分别为(1.37±0.06),(0.81±0.03)。与模型组比较,参附各剂量组心肌细胞的凋亡率明显减少,Bcl-2蛋白表达显著增加,Caspase-3蛋白表达显著减少(P<0.01)。结论:参附注射液对阿霉素引起的心肌损害有保护作用,并提示其机制可能与上调Bcl-2蛋白表达,下调Caspase-3蛋白表达相关。Objective: To study the mechanism of Shenfu injection protection of myocardial apoptosis caused by adriamycin. Methods: Totally 70 rats were randomly divided into five groups with 14 rats in each group : normal control group, adriamycin injured group and three dosage groups.The normal control group was only treated with saline by intraperitoneal injection throughout the course of the experiment, while the other four groups were given adriamycin by intraperitoneal injection with the total dosage of 20mg/kg. The different dosage of Shenfu Injection was administrated by gavage to the three treated groups.Pathological changes in cardiomyocyte were observed. Apoptosis rates were determined. The protein expressions of Bcl-2, Caspase-3 in cardiomyocytes were detected by western blot analysis. Results: Apoptosis index in the model group and the Shenfu injection group were (30.15±1.26)% and (12.74±1.03)%. The protein expressions of Bcl-2 and Caspase -3 in the model group were (0.40±0.04) and (1.37±0.01), while the protein expressions of Bcl-2 and Caspase-3 in the high -dose group were (1.37±0.06) and (0.81±0.03). Compared with those in the model group, apoptosis index in the SF group decreased significantly (P〈0.01), the expressions of Bcl-2 were higher and those of Caspase-3 were lower (P〈0.01). Conclusion:. Shenfu Injection could inhibit myocardial cell apoptosis in rats and its mechanism may be through up-regulating Bcl-2 and down-regulating Caspase-3 protein expression.
