摘 要:目的探讨1,25-二羟维生素D3[1,25(OH)2D3]对Ig A肾病大鼠调节性T细胞的作用。方法 26只Wistar大鼠分为正常对照组、Ig A肾病组、1,25(OH)2D3治疗组。检测大鼠尿蛋白定量(24 h)的变化,采用实时荧光定量聚合酶链反应检测人叉头框蛋白P3(Foxp3)m RNA的表达水平。结果 1Ig A肾病组与正常对照组相比,尿蛋白定量(24 h)水平显著升高,Foxp3 m RNA的水平显著降低,差异有统计学意义(P<0.05)。2治疗组与Ig A肾病组相比,尿蛋白定量(24 h)水平显著降低,Foxp3 m RNA的水平显著升高,差异有统计学意义(P<0.05)。结论11,25(OH)2D3可能降低或缓解Ig A肾病尿蛋白的水平。21,25(OH)2D3可能通过调节调节性T细胞分化发挥免疫作用。Objective To explore the effects of 1,25-dihydroxyvitamin D3 on regulatory T cells in rats with IgA nephropathy (IgAN). Methods Twenty-six Wistar rats were assigned to healthy control group, IgAN group, and 1,25-dihydroxyvitamin D3 treatment group. The 24-hour urine protein was measured, and the level of Foxp3 mRNA was as-sayed by real-time polymerase chain reaction. Results Compared with healthy control group, the level of 24-hour urine protein was significantly higher, and the expression of foxp3 was significantly lower in IgAN group (both P〈0.05). After treatment with 1,25-dihydroxyvitamin D3, the level of 24-hour urine protein was significantly reduced and the level of Foxp3 expression was markedly increased (both P〈0.05). Conclusion 1, 25-dihydroxyvitamin D3 might re-duce the level of urinary protein, the protective role of which on IgA nephropathy might be associated with the regula-tion of Foxp3 expression in rats with IgA nephropathy.
