摘 要:目的:观察不同电针对慢性应激抑郁模型(chronic unpredictable mild stress,CUMS)大鼠额叶、海马、下丘脑神经肽Y(NPY)表达的影响,探讨NPY在大鼠实验性抑郁症发病过程中的作用机制及不同电针干预对其的影响。方法:将60只清洁健康雄性SD大鼠,随机分为正常组、模型组、脉冲电针组、音乐电针组及氟西汀组,每组12只。孤养结合CUMS 21天复制抑郁症模型,通过开野试验观察大鼠行为学变化,采用荧光实时定量PCR检测NPY在大鼠额叶、海马、下丘脑中的表达情况。结果:模型组大鼠开野实验水平运动及垂直运动明显减少,脉冲电针组、音乐电针组及氟西汀组均可改善此变化;模型组大鼠额叶、海马、下丘脑内NPY表达均明显降低,脉冲电针组、音乐电针组及氟西汀组可逆转此病理变化,但氟西汀组治疗效果略优于两组电针治疗效果;脉冲电针效果优于音乐电针。结论:抑郁大鼠的NPY含量低于正常;电针治疗抑郁症可能是通过升高NPY的含量而发挥抗抑郁作用;脉冲电针效果优于音乐电针。Objective: To investigate the influence of different electro-acupuncture( EA) treatment on the expression of NPY in the frontal lobe,hippocampus,hypothalamus of rats with chronic unpredictable mild stress,to explone the possible role of NPY in the development of experimental depression and the effect of different EA for them. Methods: Adult male SD rats were randomly divided into normal control group,model group,pulsed EA group,music EA group and fluoxetine group. Depression model was established by lonely raising and chronic unpredictable mild stress for 21 days. Open-field test was performed to determine the behavior of rats.The expression of NPYmRNA in frontal lobe,hippocampus,hypothalamus was detected by using real-time PCR techniques respectively. Results: The cross and rear score of the model group were significantly lower than those of normal control group,while pulsed EA group,music EA group and fluoxetine group could reverse it. The expression of NPYmRNA in frontal lobe,hippocampus,hypothalamus of model group decreased significantly,the three kinds of treatment could improve these pathological change,but the effect of fluoxetine was better than that of the other two EA group; in addition,the effects of pulse-EA was better than that of music-EA treatment.Conclusion: The levels of NPY in frontal lobe,hippocampus,hypothalamus of depression rats were lower than normal. EA treatments may has antagonism against depression by increasing the levels of NPY in frontal lobe,hippocampus,hypothalamus,the effects of pulse-EA was better than that of music-EA treatment.
