安肺益肾方对慢性阻塞性肺疾病模型大鼠血清PDGF、TGF-β的影响

摘　　要：目的:探讨安肺益肾方对慢性阻塞性肺疾病模型大鼠血清PDGF、TGF-β的影响。方法:健康Wistar大鼠72只,采用半随机单盲法分为安肺益肾方大、中、小剂量组,氨茶碱组,模型组,正常对照组,每组12只,除正常对照组外,其余各组大鼠采用熏香烟加气管注入内毒素法建立慢性阻塞性肺疾病模型。安肺益肾方大、中、小剂量组分别给予生药2.4、1.2、0.6g/kg,氨茶碱组给予氨茶碱15mg/kg,模型组和正常对照组灌胃0.5%羧甲基纤维素溶液,连续给药4周。ELISA法检测血清PDGF、TGF-β含量。结果:安肺益肾方能降低慢性阻塞性肺疾病模型大鼠血清PDGF、TGF-β表达(P<0.01)。结论:安肺益肾方具有抑制慢性阻塞性肺疾病模型大鼠肺基质的生成、重构和纤维化作用。Objective： To investigate the effects of Anfei yishen formula（ AFYSF） on serum PDGF and TGF-β levels of rats with chronic obstructive pulmonary disease（ COPD）. Methods： 72 Wistar rats were divided into AFYSF high,middle,low dose groups,aminophylline group,model group,normal control group by a randomized single blind method,there were 12 rats in each group. The models with COPD were induced by intratracheal instillation of lipopolysaccharide and passive smoking,except normal control group,rats were given sodium carboxymethyl cellulose,AFYSF（ crude drugs 2. 4、1. 2、0. 6g / kg,three dosage groups） and aminophylline respectivety,each group got four weeks treatment. Then the PDGF and TGF-β levels in serum were tested with ELISA. Results： AFYSF can reduce the expressions of serum PDGF and TGF-β in rats with COPD（ P〈0. 01）.Conclusion： AFYSF could inhibit matrix formation,remodeling and fibrosis of lung in rats with chronic obstructive pulmonary disease.
