摘  要:目的:观察交泰丸对db/db小鼠的治疗作用,并通过观察其对脂肪组织PGC-1α和GLUT4基因表达的影响,探讨其部分作用机制。方法:将db/db小鼠分为模型组、交泰丸组和罗格列酮组,db/m小鼠作为正常对照组。药物灌胃治疗4周,正常组和模型组给予生理盐水。观察体重、饮水量、进食量、尿量变化,观察空腹血糖(FBG)、腹腔注射葡萄糖耐量(IPGTT)和脂肪组织PGC-1α(RT-PCR)、GLUT 4(Western blot)表达的差异。结果:与正常组比较,db/db小鼠组均呈现肥胖、多饮、多食、多尿、高血糖症状;与模型组比较,交泰丸组饮水量、摄食量、尿量、FBG、IPGTT均降低(P<0.05);PGC-1α和GLUT 4表达均增多(P<0.05)。结论:交泰丸对db/db小鼠有较好的降糖效果,降糖机制可能与增加脂肪组织PGC-1α和GLUT 4的表达有关。Objective: To study the effects of Jiaotai pill on type 2 diabetes mellitus db / db mice,explore the part mechanism by observing the influence on PGC-1α and GLUT4 gene expressions in adipose tissue. Methods: db / db mice were randomly divided into model group,Jiaotai pill group,rosiglitazone group and db / m mice were regarded as normal group. Mice in different treatment groups were given corresponding therapy for 4 weeks,mice in normal group and model group were received only physiological saline. During this period,body weight,water intake,food-intake,urine volume,fasting blood glucose( FBG),intraperitoneal injections of glucose tolerance( IPGTT),PGC-1α( RT-PCR) and GLUT4( western blot) expressions were determined after 4 weeks.Results: To compare with normal group,model group mice showed obesity,polydipsia,polyphagia,polyuria,hyperglycemia,to compare with model group,water intake,food intake,urine volume,FBG,IPGTT all had a significant decrease( P〈0. 05) in Jiaotai pill group; PGC-1α and GLUT4 expressions had a significant increase( P〈0. 05). Conclusion: Jiaotai pill is effective for the treatment of type 2 diabetes mellitus mice,the possible mechanism is connected with the increasing of PGC-1α and GLUT4 gene expressions in adipose tissue.
