小鼠脊髓损伤后Aβ蛋白的表达与细胞凋亡

摘　　要：目的：观察小鼠急性脊髓损伤后不同时间点Aβ蛋白的变化与损伤周围区神经细胞的凋亡情况,进一步了解脊髓损伤后相关的病理生理变化。方法：130只昆明（KM）小鼠随机分为模型组60只、假手术组60只、对照组10只。模型组和假手术组均在脊髓胸8-12节段打开椎板,暴露脊髓。模型组采用改良Allen法打击脊髓后缝合伤口;假手术组打开椎板暴露脊髓后直接缝合。模型组和假手术组分别于2 h、12 h、24 h、3 d、7 d、14 d六个时间点进行模型制作,脊髓损伤部位取材。对照组直接取相同脊髓节段,免疫荧光组织化学和Western Blot方法观察Aβ蛋白与细胞凋亡的变化。结果：脊髓损伤后模型组各个时间点的Aβ蛋白的含量和阳性细胞的数量较假手术组各时间点和对照组明显增加（P〈0.05）,模型组各个时间点的Aβ蛋白的含量和阳性细胞的数量随术后时间的增长而增加,3-7 d达到高峰,呈现一定的规律性。假手术组各个时间点的结果均无明显变化;细胞凋亡的百分比在脊髓损伤后随时间改变逐渐增加,模型组较对照组增加明显（P〈0.05）。结论：脊髓损伤后Aβ蛋白的表达随时间的变化呈规律性改变;细胞的凋亡具有相似的趋势。Objective：To study the expression level of Aβ protein and apoptosis in damaged spinal cord and explore the pathophysiological changes at different times after the injury.Methods：130 Kunming adult mice were randomly divided into the operation group（ 60 mice）,the sham group（ 60 mice） and the control group（ 10 mice）.The operation and sham group mice were made laminectomy on thoracic 8 - 10（ T8 - 10） to expose the spinal cord.In the operation group,spinal cord was hit to cause damage according to the improved Allen＇s method.Mice in sham group were not damaged in the spinal cord and were sutured incision after the surgery.T8-10 spinal segments in different group mice were removed and observed by immunofluorescence histochemistry and Western Blot at 2 h,12 h,24 h,3 d,7 d and 14 d after the operation.Results：After the spinal cord injury,Aβ protein content and the number of positive cells in the operation group were significantly increased compared with that in the sham and control groups at each time point（ P〈0.05）.In the operation group,Aβ protein content and the number of positive cells at each time point increased with the growth of postoperative time,reached its peak during 3-7 days after the damage and showed certain regularity.In the sham group,the results for each time point did not change significantly.Percentage of cell apoptosis after spinal cord injury gradually increased o-ver time.The trend in the operation group increased significantly compared with that in the sham group（ P〈0.05）.Conclusions：After spinal cord injury,Aβ protein expression presented a regular change with the time and cell apoptosis had a similar trend.
