摘 要:目的:探讨枸杞多糖(Lycium barbarum polysaccharide,LBP)对大鼠癫痫发作后海马齿状回颗粒细胞层神经细胞的影响。方法:选择健康5周龄雄性SD大鼠,随机分为三组,即对照组、癫痫组和LBP干预组。癫痫组腹腔注射氯化锂-匹罗卡品,LBP干预组在注射氯化锂-匹罗卡品前分别灌服25 mg/kg、50 mg/kg、100 mg/kg的LBP 2周,对照组给予等剂量生理盐水。溴脱氧尿嘧啶核苷(Brd U)标记后免疫荧光染色,观察大鼠海马齿状回颗粒细胞层Brd U阳性细胞的增殖情况。结果:与对照组相比较,癫痫组的Brd U阳性细胞明显增多(P<0.01);与癫痫组相比较,不同剂量LBP干预组的Brd U阳性细胞均明显减少(P<0.01),但各剂量LBP干预组间无显著性差异(P>0.05)。结论:LBP可干预大鼠癫痫发作后海马齿状回颗粒层Brd U阳性细胞数的异常增生,具有明显的神经保护作用。Objective:To investigate the influence of Lycium barbarum polysaccharide( LBP) on the nerve cells of hippocampal dentate gyrus granular cell layer in epilepsy rat.Methods:Male Sprague-Dawley rats( 5 weeks) were randomly assigned into three groups:control group,epilepsy group,and intervention group.Lithium-pilocarpine was injected into the abdominal cavity to create epilepsy model in rats.Intervention group rats were administered with LBP( 25 mg / kg,50 mg / kg,100 mg / kg) via oral gavage once a day for two weeks,and then injected Lithium-pilocarpine,while the control group received only same volume of saline.After the treatment,the number of 5-bromodeoxyuridine( Brd U) labeling of positive cells was observed by immunofluorescence staining.Results:The levels of Brd U positive cells are remarkably enhanced in epilepsy group compared with control group( P〈0.01).The number of Brd U positive cells in animals treated orally with LBP significantly decreased compared to epilepsy group( P〈0.01).But withdrawal of oral treatment with LBP was associated with non-significant alterations in other indicators( P〉0.05).Conclusion:LBP can decrease the abnormal proliferation of Brd U positive cells of hippocampal dentate gyrus granular cell layer in epilepsy rat.The result may support the fact that LBP has obvious nerve protective effect.
