摘  要:目的:本研究旨在观察丹酚酸B对体外培养新生大鼠海马神经元的影响,为丹酚酸B对中枢神经系统的作用提供实验依据。方法:体外培养新生大鼠海马神经元,神经元微管相关蛋白-2(MAP-2)免疫荧光染色对培养细胞进行鉴定,实验分为对照组和给药组(终浓度分别为5 mg/L和10 mg/L),荧光显微镜下观察细胞形态,MTT比色法检测丹酚酸B对海马神经细胞的活性,细胞核染色(DAPI)观察丹酚酸B对海马神经细胞增殖的影响。结果:免疫荧光染色显示细胞呈MAP-2阳性,丹酚酸B作用7 d后,能明显促进海马神经细胞的存活与增殖,且10 mg/L的丹酚酸B作用更明显。结论:丹酚酸B能明显促进海马神经细胞的存活与增殖,提示其具有改善脑功能的重要作用。Objective:To explore effect of salvianolic acid B on cultured hippocampal neurons in neonatal rat and provide experiment basis for role of salvianolic acid B on central neural system.Methods:The neonatal rat hippocampal neurons were cultured in vitro and identified by the neuronal microtubule associated protein( MAP-2) immunofluorescence labeling.The experiments were randomly divided into control group and salvianolic acid B group( 5 mg / L and 10 mg / L,respectively).The cellular morphology was observed by fluorescence microscope.The neurons viability was determined by MTT assay.DAPI staining was used to detect the proliferation of cultured neurons.Results:Immunofluorescent staining showed that the cells were MAP-2 positive.Compared with the control group,the hippcampal neurons viability and proliferation in the salvianolic acid B were significantly increased on 7 days,and the effect was more obvious with 10 mg / L salvianolic acid B.Conclusion:The salvianolic acid B can obviously promote hippocampal neurons viability and proliferation,indicating the important role of the salvianolic acid B on improving brain function.
