摘  要:目的探讨长链非编码RNA(LncRNA)在宫颈癌及宫颈上皮内瘤样病变(CIN)中的表达。方法 60例宫颈组织标本按病理诊断分为宫颈癌组、CIN组和正常组,每组20例。通过LncRNA数据库筛选在宫颈组织中可能异常表达的LncRNAs,采用实时荧光定量PCR(qRT-PCR)进行验证。结果通过NONCODE科学数据库筛选出与宫颈组织相关的LncRNA H19、MALAT1、HOTAIR、BCYRN1、ANRIL、SRA和UCA1。qRT-PCR结果:60例标本中LncRNA H19在宫颈癌组、CIN组标本中的表达量低于正常组(P<0.01);SRA在宫颈癌组中的表达量低于CIN组(P<0.01);HOTAIR、UCA1和MALAT1在CIN组中的表达量高于宫颈癌组及正常组(P<0.05);BCYRN1和ANRIL在三组中的表达量无统计学差异(P>0.05)。结论 H19在宫颈癌和CIN发生中起抑制作用;SRA可抑制宫颈癌发生,但对CIN的发生有促进作用;HOTAIR、UCA1和MALAT1促进CIN发生。LncRNAs可能成为宫颈癌及CIN早期诊断和治疗的新监测指标。Objective To investigate the expressions of long non-coding RNA(LncRNA)in cervical cancer(CC)and cervical intraepithelial neoplasia(CIN).Methods According to pathological diagnosis,60 cases were equally divided into 3 groups of CC,CIN and NC(normal cervical tissues).The possible abnormal expression of LncRNAs was screened by LncRNA database.Quantitative real time-PCR(qRT-PCR)was used to confirm the expression of LncRNAs in 60 samples.Results The LncRNAs screened by NONCODE were H19,MALAT1,HOTAIR,BCYRN1,ANRIL,SRA and UCA1.The results reported by qRT-PCR showed that LncRNA H19 expression was lower in groups of CC and CIN than that in group NC(P〈0.01),SRA expression was lower in group CC than that in group CIN(P〈0.01),the expressions of HOTAIR,UCA1 and MALAT1 were higher in group CIN than those in groups of CC and NC(P〈0.05),and the expressions of BCYRN1 and ANRIL were not significantly different among three groups(P〉0.05).Conclusion LncRNA H19 has an inhibitory effects on CC and CIN.SRA may suppress the occurrence of CC,but promote the occurrence of CIN.HOTAIR,UCA1 and MALAT1 may promote the occurrence of CIN.The LncRNAs could be new indicators for early diagnosis and gene therapy for CC and CIN.
