摘  要:目的观察通过使用不同浓度黄芩素干预宫颈癌He La细胞后,TIMP2及凋亡相关因子Bax、Bcl2m RNA及蛋白水平表达的变化。方法体外培养He La细胞株,分为空白对照组、100μmol/L干预组、200μmol/L干预组,培养24h。相差显微镜下观察细胞形态及数量的变化。流式细胞技术法检测细胞周期的变化。RT-PCR法检测TIMP2、Bax、Bcl2m RNA水平的表达变化。Western Blot法检测TIMP2、Bax、Bcl2蛋白水平的表达变化。结果黄芩素干预24 h后,与空白对照组相比较,各组细胞G1期均受阻滞,组织程度与黄芩素干预浓度成正向变化,Bcl2的m RNA及蛋白表达与黄芩素干预浓度浓度相关,呈逐渐降低的趋势(P<0.05),TIMP2、Bax的m RNA及蛋白水平表达趋势为随黄芩素浓度增加而逐渐增强(P<0.05)。结论黄芩素通过阻滞细胞周期中G1期,上调Bax,下调TIMP2和Bcl2的表达,促使He La细胞凋亡。Objective To observe the changes of mRNA and protein expression of cell tissue inhibitor of metalloproteinase-2 (TIMP2) and Bcl2-Associated X Protein (Bax) And B cell lymphoma/lewkmia-2 (Bcl2) in cervical cancer cells stimulated by baicalein. Methods HeLa cells cultured in vitro were exposed to baicalein (0, 100 and 200 μM) for 24 h. Then the periodic changes of each group cell were detected by flow cytometry, mRNA changes of TIMP2and Bax/Bcl2 were measured by PCR and protein expression of TIMP2, Bax/Bcl2 was measured by western blot. Results Compared to control group, baiealein significantly decreased the mRNA and protein expression of Bcl2 in a concentration-dependent manner (P〈0.05). The mRNA and protein expression of TIMP2 and Bax significantly increased (P〈0.05). Conclusion Baicalein induced HeLa cell growth inhibition by inhabiting cell cycle G1, and by decreasing expression of Bcl2 and increasing expression of TIMP2 and Bax.
