摘  要:目的探讨橙皮素对脂多糖(lipopolysaccharides,LPS)诱导H9C2心肌细胞活性氧(reactive oxygen species,ROS)水平的影响。方法使用不同浓度橙皮素(0.0625、0.125、0.25μmol/L)和LPS(1μg/ml)共同孵育H9C2心肌细胞120min,DCFHDA探针检测H9C2细胞ROS水平,荧光显微镜定性观察以及流式细胞仪定量检测各组细胞荧光强度,以观察不同浓度橙皮素对LPS诱导H9C2心肌细胞产生ROS的影响。选择0.25μmol/L橙皮素和LPS(1μg/ml)共同孵育H9C2心肌细胞30、60、120min,观察橙皮素对LPS诱导H9C2心肌细胞产生ROS的时间相关性。结果橙皮素干预可以浓度依赖性的缓解LPS诱导H9C2心肌细胞ROS产生;在60、120min,橙皮素能够抑制LPS诱导H9C2心肌细胞ROS产生。结论橙皮素能够抑制LPS诱导的H9C2心肌细胞ROS产生,从而降低心肌细胞氧化应激水平,其有可能成为心肌保护新的潜在药物。Objective To investigate the effect of hesperetin on the level of reactive oxygen species(ROS) in H9C2 cardiomyocytes which induced by lipopolysaccharides(LPS).Methods The embryonic rat heart-drived H9C2 cells were coincubated with different concentrations of hesperetin(0.0625,0.125,0.25 μmol/L)and LPS(1 μg/ml)for 120min.And then H9C2 cells were coincubated with hesperetin(0.25μmol/L)and LPS(1μg/ml)for different time(30,60,120min).Results We found that H9C2 cells treated with hesperetin could be protected from LPS-induced ROS production.The decreasing ROS levels in LPS-stimulated cells were concentration-dependent and time-related by hesperetin treatment.Conclusion Hesperetin can reduce the ROS production and attenuate oxidative stress in H9C2 cardiomyocytes which induced by LPS.
