摘 要:目的测定不同时期烧伤患者增生性瘢痕(HS)组织中前列腺素(PGs)和基质金属蛋白酶-2(MMP-2)的含量并探讨其变化规律。方法选取2013年3-9月在无锡市第三人民医院收治的烧伤患者或需手术切除瘢痕的患者皮肤标本57例,分为正常组(10例),烧伤早期组(9例),瘢痕1个月组(9例)、3个月组(9例)、6个月组(8例)、12个月组(6例)、24个月组(6例)。采用ELISA法和Western bolt法测定不同时期各分组瘢痕组织中PGs(包括PGD2,PGE2和PGF2α)和MMP-2含量。观察PGs和MMP-2在不同时期瘢痕组织中的含量变化。结果正常组PGE2、PGD2和PGF2α的含量最低[(5.543±0.484)、(3.619±0.484)、(4.833±1.106)ng/m L],与正常组比较,烧伤早期组PGE2、PGD2和PGF2α含量显著增加[(30.960±0.735)、(25.952±1.309)、(17.451±1.056)ng/m L](P〈0.01);除正常组外,瘢痕24个月组PGE2、PGD2和PGF2α的含量最低[(11.445±0.972)、(8.012±0.972)、(5.888±1.059)ng/m L],但与正常组比较差异仍有统计学意义(P〈0.01或P〈0.05)。MMP-2在正常组含量最低,与正常组比较,烧伤早期组MMP-2的增加(P〈0.05),瘢痕3个月组含量最高(P〈0.01),瘢痕12个月组降低(P〈0.05),瘢痕24个月组MMP-2含量则降低至与正常组接近的水平(P〉0.05)。结论 PGs和MMP-2在不同时期瘢痕组织中的含量和正常皮肤比较有显著差异,提示两者可能在增生性瘢痕中有重要作用。Objective To determine and analyze the contents of prostaglandins(PGs) and matrix metalloproteinase-2(MMP-2)of hyperplastic scar(HS) in burned patients at different stages and discuss its change regular. Methods March to September 2013, in the Third People's Hospital of Wuxi City, 57 skin samples from patiens with burn or needed surgery to remove scar were divided into different groups according to different stages, normal group(10 cases), early period burn group(9 cases), HS 1 month group(9 cases), HS 3 months group(9 cases), HS 6 months group(8 cases), HS12 months group(6 cases), and HS 24 months group(6 cases). The contents of PGs(include PGE2, PGD2 and PGF2α)were measured by ELISA, the expression of MMP-2 was determined by Western bolt. The contents changes of PGs and MMP-2 in HS patients at different stages were observed in this study. Results In normal group, the contents of PGE2,PGD2 and PGF2α were the lowest [(5.543±0.484),(3.619±0.484),(4.833±1.106) ng/m L], compared with normal group,the contents of PGE2, PGD2 and PGF2α significantly increased in early period burn group [(30.960±0.735),(25.952±1.309),(17.451±1.056) ng/m L]. After that time, the contents of PGE2, PGD2 and PGF2α showed decreasing trend. Except normal group, the contents of PGE2PGD2 and PGF2α in HS 24 months group were the minimum [(11.445±0.972),(8.012±0.972),(5.888±1.059) ng/m L], but compared with normal group, the differences were still statistically significant(P〈0.01 or P〈0.05). The expression of MMP-2 In normal group was the lowest, compared with normal group, MMP-2 expression in early period burn group were increased(P〈0.05), MMP-2 expression in HS 3 months group was the maximum(P〈0.01), that in HS 12 months group showed reduce(P〈0.05), that in HS 24 months group decreased to approximate normal level(P〉0.05).
