摘  要:为提高新城疫病毒(NDV)弱毒疫苗的免疫保护效果,本研究选取鸡IL2(chIL2)和chIL15两个细胞因子作为分子佐剂,通过反向遗传操作技术分别将这两个基因克隆于NDV Clone30株中,构建表达chIL2和chIL15的重组病毒rClone30-chIL2和rClone30-ch IL15。ELISA法检测结果表明两种细胞因子在重组病毒中均能够稳定表达。将重组病毒免疫雏鸡,以NDV强毒BJ株进行攻毒试验,测定其抗体产生情况和保护效果。结果表明,两种重组病毒均可以诱导雏鸡产生较高水平抗体;而且两种重组病毒保护率可达100%,具有较好的保护效果。本研究为进一步提高NDV重组基因工程疫苗的效果提供了新思路。To improve the protective efficacy of attenuated Newcastle disease virus (NDV) vaccine,the genes of chicken IL2 (chIL2) and chIL15 were amplified by PCR and inserted into the NDV vector of Clone30 strain to construct rClone30-chIL2 and rClone30-chIL15 by reverse genetic system.RT-PCR identifications showed that the recombinant virus of rClone30-chIL2 and rClone30-chIL15 contained the foreigntarget genes; ELISA assays indicated that the foreigntarget genes of the cytokines were able to be stably expressed in the recombinant virus infected cells.In addition,the HI antibody titers against NDV in chicken groups immunized rClone30-chIL2 and rClone30-chIL15 were 4log2 and 3.6log2 at 7 days post vaccination (pv),while Clone30 group and the control group were 3log2 and llog2,respectively.Furthermore,the recombinant viruses also provided better protections for the chickens than that of the parental virus against the challenge at 7 days pv which had potential for early protection against NDV infection.
