摘 要:目的研究50 Hz、0.1 m T三角波电磁场对大鼠峰值骨量(peak bone mass,PBM)的影响。方法将5周龄的雌性SD大鼠分成对照组和实验组(n=9)。实验组每天使用三角波电磁场处理90 min,对照组除不进行磁场干预外其他条件与实验组相同。在磁场处理中的第1、2、3个月末,腹腔麻醉测量骨密度(bone mineral density,BMD)。第3个月末心脏取血处死。ELISA试剂盒检测血清中碱性磷酸酶(bone alkaline phosphatase,BALP)、瘦素(leptin,LEP)和抗酒石酸酸性磷酸酶(tartrate resistant acid phosphatase 5b,Tracp 5b)含量,并进行骨形态计量分析。结果实验组在1、2个月末,BMD检测与对照组相比无显著性差异(P>0.05);3个月末,实验组BMD检测与对照组相比有显著性差异(P<0.05)。血清生化检测结果显示实验组ALP含量上升,Tracp 5b和LEP含量下降,与对照组相比具有显著性差异(P<0.05)。实验组双荧光间距离和骨小梁宽度均高于对照组,有显著性差异(P<0.05)。结论低频三角波电磁场能够提高大鼠PBM。Objective To determine the effect of triangle wave electromagnetic field( EMF) on the peak bone mass in rats. Methods Female SD rats( 5 weeks old) were randomly divided into 2 groups,control( n = 9) and experimental group( n = 9). The rats of the latter group were exposed to a triangle wave EMF at a frequency of 50 Hz and an intensity of 0. 1 m T for 90 min every day,while those of the control group were under the same conditions as the experimental group except no magnetic field intervention. In 1,2 and 3months after magnetic treatment,bone mineral density was determined after abdomen anesthesia. All the rats were executed by drawing-out all the blood in their hearts in 3 months after treatment. ELISA was used to measure the serum contents of alkaline phosphatase( BALP), leptin( LEP) and tartrate resistant acid phosphatase( Tracp-5b). A computer-aided image processing system was employed to analyze the morphology of the femur after Van Gieson staining. Results No significant difference was found in bone mineral density between the experimental group and control in 1 and 2 months of magnetic intervention( P 〉 0. 05). But in 3months,significant difference was seen( P 〈 0. 05). Serum biochemical results showed that the serum content of ALP was increased,and those of Tracp-5b and LEP were decreased,with significant difference when compared with the control( P 〈 0. 05). The distance of the double fluorescence and the average width of bone trabecula were significantly larger in the experimental group than the control( P 〈 0. 05). Conclusion Low frequency triangular wave electromagnetic field improves the peak bone mass in rats.
