摘  要:雄性成年昆明小鼠分为亚硝酸盐染毒组和生理盐水对照组,通过灌胃进行亚硝酸盐染毒,用BrdU腹腔注射和免疫组织化学方法研究齿状回神经干细胞的增殖和新生细胞的存活状况,用多重免疫荧光标记研究神经干细胞的分化和迁移方向。结果显示,染毒组小鼠齿状回颗粒层中的BrdU阳性细胞数量、存活率均极显著低于对照组(P<0.01),表明亚硝酸盐中毒对小鼠新生神经干细胞的增殖和存活产生了影响。染毒组BrdU/NeuN双阳性细胞的数量极显著低于对照组(P<0.01)。与对照组相比,染毒组中神经干细胞的迁移方向无显著性差异(P>0.05)。结果表明,亚硝酸盐中毒能显著抑制小鼠神经干细胞增殖,降低新生细胞的存活率。To investigate the influence of nitrite poisoning on the proliferation, differentiation and survival of adult neural stem cells in mouse dentate gyrus. Healthy adult male Kunming mice were divided into control group and experimental poisoning group, the latter was exposured to nitrite by intraperitoneal injection. Immunohistochemistry was used to investigate the proliferation of adult neuraistem cells and survival of newly generated cells in the dentate gyrus of mice. Multiple fluo- rescent immunostaining was used to investigate the differentiation and migration direction of new born cells. Our results showed that both the number of BrdU-positive cells and the survival rate of new born cells in experimentally poisoning mice were remarkably decreased compared with the control group(P〈0.01) ,indicating that the differentiation and survival of adult neural stem cells were significantly influenced by the nitrite poisoning. The BrdU/NeuN double labeled cells in exper- imentally poisoning group were significantly lower than that in control group(P〈0.01). But the migra- tion direction of new born cells was not influenced by nitrite(P〉0.05). These results suggest that nitrite poisoning suppressed the proliferation of adult neural stem cells and reduced the survival rate of newly generated cells in the SGZ of the dentate gyrus in mice.
