摘 要:从神经-内分泌-免疫网络与机体免疫稳态调节的角度出发,通过建立大鼠炎症性肠病(IBD)动物模型,应用荧光定量PCR方法检测μ型阿片肽受体(MOR)在IBD大鼠不同组织和外周血淋巴细胞中的表达水平,并采用ELISA方法测定血清和脑脊液中MOR的内源性配体(β-内啡肽;强啡肽)在IBD大鼠不同发病时期的表达水平变化,探讨MOR在炎症性肠病中的调节作用。结果显示,MOR在大鼠下丘脑、胸腺和外周血淋巴细胞中的mRNA水平随着炎症的加剧而逐渐增加,而在结肠组织中随着炎症的加剧其表达逐渐降低,且在病变最为严重时变化最为显著,随着炎症反应的消退逐渐恢复至正常水平。在血清和脑脊液中,内源性配体强啡肽和β-内啡肽蛋白浓度在病变高峰期显著降低。结果表明,MOR可能在IBD发生、发展中发挥调控作用。In order to investigate the regulatory role of MOR in inflammatory process of IBD. Tri- nitrobenzenesulfonic acid-induced (TNBS) colitis was induced in rats. Colitis was assessed by clin- ical score, histological score and myeloperoxidase activity, the level of MOR in different tissues and peripheral blood lymphocytes was detected by real-time RT-PCR, and the concentration of MOR endogenous ligand (β-endorphin; dynorphin) in serum and cerebrospinal fluid were meas- ured by ELISA during the different time points after induction by TNBS. Results showed that the high mRNA levels of MOR were detected in the rat hypothalamus, thymus and peripheral blood lymphocytes with the progression of IBD, and the lowest mRNA level of MOR was detected in the colon tissue. MOR changed markedly during day 3 to 7 after induction by TNBS. Conversely,with the recovery of enteropathy and alleviation of symptoms,their mRNA levels began to change and gradually trended towards normal levels. Moreover, the protein levels of endogenous ligand dynor- phin and β-endorphin in serum and cerebrospinal fluid were significantly reduced with recovery of IBD. These results suggest that MOR may play a regulatory role in the process of the occurrence and development of IBD.
