摘 要:目的研究清脉饮及其拆方对动脉硬化闭塞症兔主动脉抑制性核因子κB(I-κB)、核因子κB(NF-κB)的mRNA表达的影响。方法选取SPF级雄性健康新西兰白兔为研究对象,通过高脂饮食、免疫损伤和右股动脉不全结扎建立动脉粥样硬化闭塞症(ASO)动物模型,36只白兔随机分为全方组、软坚组、活血组、清法组、模型对照组和空白对照组。造模的各组在造模完成后恢复普通饮食并分别予以相应药物干预8周后取材。以实时荧光定量PCR法(QPCR)检测主动脉内皮组织I-κB mRNA、NF-κB mRNA表达量的变化。结果全方组、软坚组、活血组三组雄兔I-κB mRNA表达高于模型对照组(P<0.01),与空白对照组比较差异无统计学意义(P>0.05);清法组I-κB mRNA表达高于模型对照组(P<0.01)和空白对照组(P<0.05);清法组I-κBmRNA表达高于全方组、软坚组、活血组(P均<0.01)。软坚组和清法组的NF-κB mRNA表达低于模型对照组,而高于空白对照组(P均<0.01),全方组和活血组的NF-κB mRNA表达高于空白对照组(P<0.01),与模型对照组比较差异无统计学意义(P>0.05)。结论清脉饮及其拆方可通过上调I-κB mRNA的表达和下调NF-κB mRNA的表达而发挥抗ASO的作用;清法组效果最佳。Objective To explore the effect of Qingmaiyin and its deco aaposed formulas on expression of I - κB and NF - κB in aorta of ASO rabbits. Methods ASO models were lade by means of high fat diet, immu- nologic injury, and incomplete right femoral artery ligation on 36 SPF male rabbits. These rabbits were put in- to different groups randomly. The groups contained Quanfang, Ruanjian, Huoxue, Qingfa, model control and no - treatment control. Each group, after the models were made, were fed by food, and medicine lasting for 8 weeks and then were killed and cut. The expression of I - κB mRNA and NF - κB mRNA in endothelial tissue of artery was observed by PCR. Results The expression of I - s:B mRNA among the Quanfang group, Ruan- jian group, Huoxue group was signficantly different from that in model ~;roup (P 〈0.01 ), but was not differ-ent from that in control group(P 〉0.05 ). The expression of I - κB mRNA of Qingfa group was higher than those in the control group and model group( P 〈0.05 ). The expression of Qingfa group was significantly high- er than those in Quanfang group, Ruanjian group and Huoxue group (P 〈0.01 ). In the expression of NF - κB mRNA, the Ruanjian group and Qingfa groups showed significantly lower level than the control of model group, while it showed significantly higher level than the control group ( P 〈0.01 ). The expression of Quan- fang group and Huoxue group were significantly higher than that of the control group ( P 〈 0.01 ). Conclusion Qingmaiyin and its decomposed Formulas play the role of anti ASO by upregulating the expression of I - κB mRNA and downregulating the expression of NF - κB mRNA.
