GM6001对涎腺腺样囊性癌细胞粘附、侵袭及迁移能力的影响

摘  要:目的 研究GM6001对涎腺腺样囊性癌(salivary adenoid cystic carcinoma,SACC)肺高转移性细胞(ade-noid cystic carcinoma-M,ACC-M)和肺低转移性细胞(Adenoid cystic carcinoma-2,ACC-2)的粘附、侵袭及迁移能力的影响.方法 选取处于对数期生长的ACC-M和ACC-2细胞,应用MTT法测定不同浓度的GM6001(10、15、20、25 μmol·L-)对ACC-M和ACC-2细胞干预前后粘附能力的影响;应用Transwell小室通过平均穿膜细胞数观察不同浓度的GM6001对ACC-M和ACC-2细胞干预前后侵袭及迁移能力的影响.结果 GM6001在10~25μmol·L-的范围内可抑制ACC-M与ACC-2细胞的粘附能力,ACC-M细胞粘附实验的OD值从(0.226±0.196)降低至(0.065±0.010);ACC-2细胞的(0.265±0.026)降低至(0.122±0.007).GM6001浓度在0~25 μmo·lL-的范围内干预ACC-M与ACC-2细胞后侵袭至小室下的细胞数分别从(42.00±2.550)个降至(20.40±3.435)个、(40.20±3.033)个降至(22.00±2.550)个;迁移至小室下的细胞数分别从(62.20±3.768)个降至(17.60±3.847)个、(55.40±2.302)个降至(18.20±2.387)个,GM6001对ACC-M细胞粘附、侵袭及迁移的抑制能力强于对ACC-2细胞的抑制能力.结论 GM6001能降低ACC-M及ACC-2细胞的粘附、侵袭及迁移能力,其在SACC临床治疗中可能具有应用前景.Objective To investigate the effects of GM6001 on the SACC of high pulmonary metastatic cell line ACC - M and low metastatic lung cell line ACC - 2 of adhesion, invasion and migration introduced. Methods The logarithmic phase cells of ACC - M and ACC - 2 were selected and the effects of different con- centration of GM6001 ( 10,15,20,25) μmol · L - 1 ) on the ACC - M and ACC - 2 cellg adhesion were analyzed by MTI" essay. The Transwell chamber was used to observe the influence of GM6001 in the invasion and mi- gration. Results GM6001 could inhibit the adhesion ability of ACC - M and ACC - 2 cell in the range of10- 25 μmol · L-L. The ACC -M cells adhesion inhibition of OD value was from (0.226± 0. 196)reduces to (0. 065 ±0. 010). The ACC -2 cells adhesion inhibition of OD value was from (0.265±0. 026) reduces to (0. 122 ±0. 007). The Concentration of GM6001 in the range of 0 μmol · L^-1 intervention ACC -M and ACC - 2 cells invaded to the small room next to the number of cells were (42.00 ± 2. 550) - ( 20.40 ± 3.435), (40.20 ±3. 033 ) -(22. 00 ±2. 550). Migrated to a small room next to the number of cells were (62.20±3.768)-(17.60±3.847), (55.40±2.302) -(18.20±2.387). Conclusion GM6001 has the capability to restrain ACC -M cells and ACC -2 cells from adherinlg, invasion and migration and it may has application prospect in clinic treatment.
