摘 要:以葡萄二倍体品种无核红宝石与四倍体品种香悦为亲本进行正反杂交,以杂交胚珠为试材进行胚挽救试验,研究不同取样时期对胚珠萌发率的影响,采用流式细胞仪法与染色体计数法对杂交后代幼苗倍性进行鉴定。结果表明:无核红宝石×香悦于花后60d接种胚珠萌发率最高,为8.33%,获得杂种胚挽救幼苗8株,其中有1株三倍体植株(12.5%);香悦×无核红宝石在花后83d接种胚珠萌发率最高,为29.38%,获得杂种胚挽救幼苗56株,其中有16株三倍体植株(27.59%)。In this study, the impact of inoculating stage on ovule germinating rate was investigated using Hybrid ovules from the reciprocal crosses between diploid seedless cultivar "Ruby Seedless" and tetraploid cultivar "Xiangyue'. Furthermore, the chromosome ploidy of these hybrids was determined by flow cytometry and chromosome counting. The main results were as follows. The highest ovule germinating rate (8.33%) appeared when inoculation was done 60d after flowering for the cross "Ruby Seedless" x "Xiangyue', from which 8 seedlings were rescued, among which one seedling was triploid (triploid percentage 12.5%). For the cross "Xiangyue" x "Ruby Seedless', the highest germination rate (29.38%) was obtained when inoculation was done 83d after flowering. 56 seedlings were rescued from this cross, and 16 of them were triploid (27.59%).
