摘 要:目的建立同时测定苦豆子药材中金雀花碱、苦参碱、氧化苦参碱、槐果碱、氧化槐果碱和槐定碱的RP-HPLC方法,测定不同产地苦豆子药材中不同部位6种生物碱的含量。方法色谱柱为Kromasil C18(4.6 mm×250 mm,5μm);流动相为甲醇(A)-0.2%磷酸水(B);洗脱梯度为:0 min,3%A;20 min,5%A;40 min,6%A;60 min,7%A;紫外检测波长为215 nm,流速为1 m L·min-1,柱温30℃。结果 6种生物碱的线性关系良好(r≥0.999 3),平均回收率为97%~104%。各产地苦豆子样品药材中果实部位的6种生物碱含量都明显高于根、茎、叶、壳部位中各碱的含量;宁夏、甘肃的药材果实部位中主要生物碱含量稍高于其他地区。结论该方法简便、准确、快速,适用于苦豆子生物碱的含量测定。OBJECTIVE To establish a method for simuhaneous determination of six alkaloid components of Sophora alopecuroides L, including cytisine,matrine, oxymatrine,sophocarpine, oxysophocarpine, and sophoridine by HPLC and determine the contents of the six alkaloids in different parts of Sophora alopecuroides L from four different areas. METHODS The analysis was carried out with Kromasil C18 chromatographic column(4. 6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol (A) and 0. 2% phosphoric acid in water (B). The gradient elution program was as follows: 0 min, 3% A; 20 min, 5% A; 40 min, 6% A; 60 min, 7% A. The UV detection wavelength was set at 215 nm. The flow rate was 1 mL · min^-1 , and the column temperature was maintained at 30 ℃. RESULTS The six alkaloids all showed good linearity (r ≥ 0. 999 3 ). The average recoveries were in the range of 97% to 104%. The contents of the six kinds of alkaloids in the fruits of all the samples were significantly higher than in other parts of Sophora alopecuroides L. The contents of the six alkaloids in the fruits of the samples from Ningxia and Gansu were slightly higher than those from other areas. CONCLUSION The method is simple and accurate with good repeatability. It is suitable for determining the contents of alkaloids in Sophora alopecuroides L.
