摘 要:目的优化琼枝麒麟菜多糖(EGP)的超声提取方法,并探讨EGP在皮肤损伤修复方面的作用。方法采用单因素考察和正交试验方法优化超声方法提取琼枝麒麟菜多糖(EGP),苯酚-浓硫酸法测定EGP中多糖含量。通过MTT细胞毒性实验、细胞增殖实验和划痕实验,研究EGP对人永生化表皮细胞(HaCaT)形态、增殖分化和细胞迁移速度的影响,评价其在皮肤损伤修复方面的作用。结果确定EGP的最优超声提取条件为:温度60℃,料液比1∶100,时间30min,EGP得率为40.3%±1.8%,其中多糖含量为48.2%±4.6%。EGP对HaCaT细胞几乎无毒,且在一定浓度范围内对HaCaT细胞的增殖有显著的促进作用;EGP还能加快HaCaT细胞的迁移速度,对皮肤损伤的愈合有显著的影响意义。结论超声法提取EGP时间短,工艺稳定,所得EGP毒性小,对皮肤及黏膜损伤的修复具有促进作用,可广泛应用于化妆品及医疗器械。Objective To optimize the ultrasonic extraction process of Eucheuma gelatinae polysaecharides (EGP) and evaluate the EGP's repair and protection activities. Methods The ultrasonic extraction process of EGP was optimized by single factor study and orthogonal experiment design, and the content of polysaccharide was measured using phenol-sulfuric acid method. The EGPs repair and protection activities were also evaluated in terms of cell morphology, cell growth and cell migration on HaCaT cell line. Results The results showed that the optimized extraction process was at the following condition: 60℃, solid-liquid ratio of 1 :100, and 30 rain, and EGP yielded 40. 3%± 1.8% which contented 48. 2%±4. 6% of polysaccharide. Cell proliferation and migration assays demonstrated that EGP played an important role to facilitate the HaCaT cell proliferation and migration with no toxicity, which might accelerate skin repair and wound healing. Conclusion It was an energy saving and stable technology to extract EGP by ultrasound. The extractive could improve the wound healing and regeneration of skin and mucosa membrane with no toxicity. EGP showed great potential in the fields of cosmetics and therapeutic devices.
