摘  要:目的分析华支睾吸虫感染小鼠肝脏中Nrf2和Trx1编码基因的表达情况,初步探讨Nrf2及Trx1在华支睾吸虫致病过程中的作用。方法选用健康Balb/c小鼠建立华支睾吸虫感染模型,分别在实验第0、3、5、7、10、14、21、28和35d摘除小鼠眼球采血,分离血清,检测血清中天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)活性;颈椎脱臼处死小鼠取肝脏,采用逆转录聚合酶链反应(RT-PCR)检测肝脏中Nrf2和Trx1的m RNA表达情况。结果与对照组相比,感染组小鼠在感染第5、7、10、14、21和35d血清ALT活性显著升高(P<0.05);分别为(19.9332±1.98180)U/L、(64.3699±14.53874)U/L、(46.7455±6.27572)U/L、(99.0038±25.34329)U/L、(46.7590±8.85478)U/L和(50.3374±14.19886)U/L。感染组小鼠抗氧化应激通路上游基因Nrf2和下游Trx1的m RNA表达升高;其中Nrf2在感染第5天升高显著(P<0.05);Trx1在感染第21d升高显著(P<0.05)。结论 Nrf2和Trx1 m RNA在华支睾吸虫感染过程中表达升高,提示Nrf2/Trx1通路可能在华支睾吸虫致病过程中发挥一定的作用。Objective To analyze Nrf2 and Trxl gene expression in mouse liver and preliminarily investigate the role ofNrf2 and Trxl in pathogenic process of Clonorehis sinensis. Method Healthy Balbic mice were selected and infected withClonorchis sinensis to establish infection model. In the infection of 0,3rd day, 5th day, 7th day, 10th day, 14th day, 21st day,28th day, and 35th day, mice blood were respectively drawn by removing the eyeballs of mice and serum was separated todetect the activity of aspartate aminotransferase(AST) and alanine aminotransferase(ALT). Then, mice were sacrificed bycervical dislocation and liver tissue was obtained. The expressions of Nrf2 and Trxl mRNA in liver of mice were detected byreverse transeriptase polymerase chain reaction(RT-PCR). Results Compared with the normal group, the activity of ALT inserum were increased (P 〈0.05) on 5th ,7th ,10th ,14th , 21st day, and 35th day of infection were (19.9332± 1.98180),(64.3699± 14.53874), (46.7455±6.27572), (99.0038±25.34329), (46.7590±8.85478) and (50.3374± 14.19886), respectively. Theexpressions of Nrf2 and Trxl mRNA were increased after infegtion, and increased significantly on 5th day and 21st day ofinfection (P 〈0.05), respectively. Conclusion The increase expressions of Nrf2 and Trxl mRNA in mice infected withClonorcbis sinensis indicated that Nrf2/Trxlpathway might play a role in pathogenic process of Clonorchis sinensis infection.
