摘 要:目的 为控制望江南(茳芒决明)药材的质量,建立望江南(茳芒决明)的显微鉴别、薄层鉴别、和含量测定标准。方法 采用粉末进行显微鉴别;采用TLC法对大黄素甲醚成分进行薄层鉴别;采用HPLC法测定茳芒决明中大黄素甲醚含量,色谱条件:色谱柱为XBridge C18(5μm,4.6 mm×250 mm),流动相为甲醇-0.1%磷酸溶液(75∶25),柱温为35℃,流速为1.0 m L/min,检测波长254 nm。结果 粉末的显微特征与文献记载基本一致。薄层鉴别色谱斑点清晰,分离度较好。含量测定中大黄素甲醚在1.942 4--24.28μg/m L之间呈良好线性关系(r=1.0),平均回收率为100.1%(n=6)。结论 本文采用的方法操作简便,准确,重复性好,可用于控制望江南(茳芒决明)药材的质量。Objective To control the quality of root tuber of Senna occidentalis var. ( L. ) Link var. sophera ( L. ) X. Y. Zhu by establishing standards of transverse section, powder, TLC and assay. Methods The powder was used to study its microscopic characteristics. TLC was adopted to analyze the physcion. The content of physcion was determined by HPLC. The XBridge C18(5μm, 4.6 mm×250 mm) column was used, the mobile phase was methanol-0.1% phosphate (75: 25), the column temperature was 35℃, the flow rate was 1.0 mL/min, and the detection wavelength was at 254 nm. Results Microscopic characteristics of its powder were in good accordance with literatures. The TLC showed the spots developed clearly with good separation. The concentration of physcion showed a good linear relationship in the range of 1. 942 4-24.28 μg/mL (r = 1.0), and the average recovery was 100.1% (RSD = 1.4%, n = 6).Conclusion The method is simple, accurate with good reproducibility, and it can be used for the quality control of roots of Senna occidentalis var. ( L. ) Link var.sophera( L. ) X. Y. Zhu.
