摘 要:目的通过整体动物实验考察菖葛益智方对血管性认知障碍(VCI)的作用机制及疗效。方法利用VCI大鼠模型,分为模型组,假手术组,菖葛益智方大、中、小剂量组共5组,观察各组模型大鼠的行为学,并检测脑组织中伊文氏兰(EB)的渗出量定量估计血脑屏障的通透性的变化。结果菖葛益智方大、中、小剂量组对模型大鼠的行为学改变相对模型组有显著改善,大、中剂量组与模型组比较,P<0.01,小剂量组与模型组比较,P<0.05;大、中、小剂量组对大鼠行为学改变均不及假手术组。小、中剂量组升高模型大鼠的脑组织EB含量,大、中剂量组对模型大鼠的脑组织EB含量相对模型组有统计学意义(P<0.05),菖葛益智方大剂量组脑组织EB含量对模型组组间有统计学意义(P<0.01);菖葛益智方大、中剂量组脑组织EB含量对假手术组有统计学意义(P<0.05),菖葛益智方小剂量组脑组织EB含量对假手术组无统计学意义(P>0.05)。结论菖葛益智方能明显改善VCI模型大鼠的行为学和病理学。Objective Through the whole animal experiment to study the effect and mechanism of Chang Ge Yizhi prescrip- tion on vascular cognitive impairment. Methods Using the model of VCI rats, divided into five groups: the model group, sham operation group, Chang C,e Yizhi Decoction in large, medium and small dose groups. Observing each group's behavior of model rats, and to detect the expression of Evans blue (EB) leakage quantitative estimate of blood brain barrier permeability changes. Results Chang Ge Yizhi prescription in large, medium and small dose group to the behavior change of model rat than model group had significant improvement. Chang C,e Yizhi Decoction high, middle dose group compared with model group there are differences( P 〈 0.01), Chang C,e Yizhi Decoction low dose group compared with model group were signifi- cantly difference( P 〈 0.05) ; Chang C-e Yizhi Decoction in large,small dose group,on the behavior of rats in sham operation group were not changed. Chang Ge Yizhi Decoction small, middle dose group of rats brain EB increased. Chang C,e Yizhi De- coction high, middle dose group of rats brain EB was relatively model group have significant difference ( P 〈 0.05 ), Chang Ge Yizhi Decoction in brain tissue dose groups of Evans blue(EB) was a very significant difference in model group ( P 〈 0. 01). Chang C,e Yizhi Decoction high, middle dose group of brain EB has the remarkable difference to the sham operation group ( P 〈 O. 05), Chang Ge Yizhi Decoction in brain tissue of small dose group of EB there were no significant differences in the sham operation group ( P 〉 0.05). Conclusion Chang Ge Yizhi Decoctiong can obviously improve the VCI behavior- istic and pathology of model rats.
