摘  要:目的:建立测定白花蛇舌草中槲皮素-3-O-β-d-吡喃葡萄糖苷含量的方法。方法:采用高效液相色谱法。色谱柱为Kromasil C18(250 mm×4.6 mm,5μm),流动相为乙腈-1%冰醋酸(60∶40,V/V),检测波长为254 nm,流速为1.0 ml/min,柱温为35℃。结果:槲皮素-3-O-β-d-吡喃葡萄糖苷的质量浓度在5.24~52.40μg/ml范围内与其峰面积积分值呈良好线性关系(r=0.999 9);精密度、稳定性、重复性试验的RSD均〈1.97%;平均加样回收率为99.57%,RSD=1.28%(n=6)。结论:该方法操作简便,结果准确、可靠,可用于白花蛇舌草药材中黄酮类活性成分的含量测定。OBJECTIVE: To develop a method for the content determination of quercetin-3-O-β-d-glucopyranoside in Hedyotis diffusa. METHODS: HPLC method was adopted. The determination was performed on kromasil C18(250 mm×4.6 mm, 5 μm)col- umn with mobile phase consisted of acetonitrile-0.1% acetic acid (60:40, V/V) at the flow rate of 1.0 ml/min. The detection wave- length was set at 254 nm, and column temperature was 35 ℃. RESULTS: The linear range of quercetin-3-O-β-d-glucopyranoside were 5.24-52.40 μg/ml (r=0.999 9) with an average recovery of 99.57% (RSD=1.28% ,n=6) ; RSDs of precision, stability and reproducibility tests were all lower than 1.97%. CONCLUSIONS: The method is simple, accurate and reliable, and can be used for the quality control of H. diffusa.
