植物抗菌组培中抗菌剂应用的研究方法

摘　　要：本文以自行研发抗菌剂组合为例介绍抗菌组培(也称开放组培)中抗菌剂应用的研究方法。该抗菌剂与MS培养基组分、植物生长调节物质有较好相容性。抗菌剂使用的推荐浓度依具体情况而异,一定的培养基分装量,培养瓶容积越大,抗菌剂的剂量也越大,夏天使用剂量比冬天高得多,培养基添加生香蕉泥,抗菌剂使用量大幅增加。该抗菌剂对油麦菜种子萌发生长、苋菜和蝴蝶兰不定芽壮苗生根、紫花苜蓿体细胞胚的壮苗生根没有明显抑制作用。本文还就抗菌组培的概念、影响抗菌剂使用适宜浓度的因素、抗菌组培在植物组培不同阶段的效用和将来研究方向等问题进行讨论。Research methods about application of antimicrobial agent in plant tissue culture with antimicrobial,or open tissue culture, are introducted with the exemples of self developed antimicrobial agents in this paper. The results of experiment showed that antimicrobial agents had compatibility with MS medium components and some plant growth regulators. The recommended concentrations of antimicrobial agents supplemented in medium were different because of the concrete circumstances. A certain loading amount of medium, the bigger culture bottle volume, the more antimicrobial agents it required. The propriate dose of antimicrobial agents in medium was much higher in summer than that in winter. The application amount of antimicrobial agents in medium that added with mashed banana would increase significantly. The antimicrobial agents in medium did not inhibite effectively germination of Lactuca sativa seed, as well as growth and rooting of Amaranthus mangostanus adventitious bud, Phalaenopsis ssp. adventitious bud and Medicago sativa somatic embryo. These are discussed in the paper, including the concept of tissue culture with antimicrobial, factors which effect on the usage dose of antimicrobial agents in medium, the role of tissue culture with antimicrobial in different stage of plant tissue culture and and some problems that will be studied.
