苦参素注射液联合顺铂对人肝癌SMMC-7721细胞端粒酶活性及wtp53、hTERT mRNA表达的影响  被引量:2

摘  要:目的:观察苦参素注射液联合顺铂(DDP)对人肝癌SMMC-7721细胞端粒酶活性、野生型p53(wtp53)、端粒酶逆转录酶(h TERT)m RNA表达的影响。方法:将Dulbecco改良Eagle培养基培养的人肝癌SMMC-7721细胞分为苦参素组、DDP组、联合组和对照组。苦参素组低、中、高质量浓度分别为0.75、1.5、3.0 mg/ml,DDP组低、中、高质量浓度分别为1、2、4 mg/ml,联合组浓度按两药联合用药1∶1的比例应用,对照组加入200μl磷酸缓冲盐溶液(PBS),不给予药物干预。观察4组人肝癌SMMC-7721细胞凋亡率及药物间的相互作用,端粒酶活性吸光度差值(ΔA),wtp53和h TERTm RNA相对表达量。结果:联合组人肝癌SMMC-7721细胞凋亡率>苦参素组>DDP组>对照组,4组比较差异有统计学意义(P<0.01),q值提示苦参素与DDP存在单纯相加或协同作用。苦参素组、DDP组及联合组SMMC-7721细胞端粒酶活性ΔA、h TERTm RNA均显著低于对照组,且联合组<DDP组<苦参素组,苦参素组、DDP组及联合组wtp53显著高于对照组,且联合组>DDP组>苦参素组,差异均有统计学意义(P<0.05)。结论:苦参素注射液联合DDP可增加人肝癌SMMC-7721细胞的凋亡,其机制可能与wtp53表达上调、h TERTm RNA表达下调,从而影响端粒酶活性有关。OBJECTIVE:To investigate the effects of Kushenin injection combined with cisplatin(DDP)on the activity of human liver cancer SMMC-7721 cells telomerase and m RNA expression of wild-type p53(wtp53)and telomerase reverse transcriptase(h TERT). METHODS:Human liver cancer SMMC-7721 cells cultured with Dulbecco modified Eagle medium were divided into kushenin group,DDP group,combination group and control group. Kushenin group was given low,medium and high concentrations of kushenin 0.75,1.5 and 3.0 mg/ml,DDP group was given 3 kinds of concentrations of DDP 1,2 and 4 mg/ml,and combination group was given two-drug combination with ratio of 1 ∶ 1;control group received 200 mu L phosphate buffer solution(PBS)without any drug intervention. The apoptosis rate of human liver cancer SMMC-7721 cells and drug interaction were observed,and telomerase activity absorbance difference(delta A),relative expression of wtp53 and h TERT m RNA were compared. RESULTS:The apoptosis rate was in descending order:combination group〉kushenin group〉DDP group〉control group,there was statistical significance(P〈0.01). q value indicated that kushenin was simply combined with or cooperated with DDP. ΔA and m RNA expression of h TERT of SMMC-7721 cells in kushenin group,DDP group and combination group were significantly lower than in control group,in ascending order as following:combination group〈DDP group〈kushenin group. m RNA expression of wtp53 in kushenin group,DDP group and combination group were significantly higher than in control group,in descending order as following:combination group〉DDP group〉kushenin group;there was statistical significance(P〈0.05). CONCLUSIONS:Kushenin injection combined with DDP can enhance human liver cancer SMMC-7721 cells apoptosis,the mechanism of which may be related that the up-regulation of wtp53 expression and the down-regulation of h TERTm RNA expression affect the telomerase activity.
