摘  要:目的建立HPLC法测定不同产地的野生槐花花蕾中芦丁与槲皮素的量。方法采用碱水提取法提取槐花花蕾中的芦丁和槲皮素。采用Agilient HC-C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.4%磷酸水溶液(52∶48)为流动相,体积流量为0.9 m L/min;检测波长为360 nm;柱温为25℃。结果芦丁在2.5-80μg/m L范围内,槲皮素在0.625-20μg/m L范围内线性关系良好;平均回收率分别为100.2%和99.1%;RSD分别为2.1%和1.1%(n=6)。来自10个产地的槐花花蕾中芦丁的量在21.48%-28.21%之间,槲皮素的量在1.15%-1.74%之间。结论不同产地槐花花蕾中芦丁和槲皮素存在一定差异,其中以河南新郑的槐花花蕾中芦丁与槲皮素含有量最高。AIM To establish an HPLC method to determine rutin and quercetin in flower buds of wild Sophora japonica L. from eight habitats. METHODS Rutin and quercetin of flower buds of S. Japonica were extracted by alkaline solution,and were determined by HPLC method. Chromatography was performed on an Agilient HC-C18( 4. 6 mm × 250 mm,5 μm) at 25 ℃,and mobile phase was composed of methanol-0. 4% phosphoric acid solution( 52 ∶ 48) with a flow rate of 0. 9 m L/min. The detection wavelength was set at 360 nm. RESULTS The linearity of rutin and quercetin were 2. 5- 80 μg / m L and 0. 625- 20 μg / m L,respectively,the average recoveries were100. 2% and 99. 1%,and their RSD were 2. 1% and 1. 1%( n = 6),respectively. The rutin content ranged from21. 48% to 28. 21%,while quercetin content ranged from 1. 15% to 1. 74%. CONCLUSION There are some differences in the contents of rutin and quercetin in flower buds of S. japonica in different habitats. Of these habitats the contents of rutin and querectin in the flower buds from Henan province are the highest.
