摘 要:目的观察乌梅黄连复方对人结肠癌细胞株HT29细胞增殖和迁移的影响。方法采用乌梅黄连复方对体外培养结肠癌细胞株HT29进行干预,MTT法检测复方对结肠癌细胞株增殖的影响,采用流式细胞术检测细胞凋亡和细胞周期,划痕试验观察复方对结肠癌细胞迁移能力的影响,采用Real-time PCR测定药物干预后细胞cox-2mRNA表达。结果乌梅黄连复方对HT29细胞具有明显的抗增殖作用,IC50为(4.55±0.25)mg/mL;抑制TPA诱导的HT29细胞的迁移(P<0.01);该复方能显著诱导HT29细胞凋亡,引起HT29细胞G2/M期阻滞,显著下调HT29细胞中cox-2mRNA的表达(P<0.05)。结论乌梅黄连方可能通过诱导肿瘤细胞凋亡,引起肿瘤细胞G2/M期阻滞,抑制肿瘤细胞cox-2通路对人结肠癌HT29细胞增殖和迁移产生抑制作用。OBJECTIVE To observe the effect of the Wumei Huanglian Fang on HT29 proliferation and migration of human colon cancer cell.METHODS Interfere the cultivation of colon cancer cell lines HT29 in vitro adopting Wumei Huanglian Fang and detect its effect on colon cancer cell line proliferation by MTT method,The (FCM) flow cytometry to detect the cell apoptosis and cell cycle was adopted,observe the effect of the formula on cancer cell migration ability by scarification test and measure the cell cox-2 mRNA express after pharmacological intervention with Real-time PCR was obseved.RESULTS Wumei Huanglian Fang had obvious antiproliferative effect on HT29 cell (IC50 =(4.55±0.25) mg/mL) ; it could inhabit the migration HT29 cell induced by TPA.This formula could significantly induce HT29 cell apoptosis,cause G2/M phase arrest of HT29 cell and significantly reduce the cox-2 mRNA expression of HT29 cell.CONCLUSION Wumei Huanglian Fang may lead to G2/M phase arrest of cancer cell by inducing tumor cell apoptosis and inhabit the inhibiting effect of cancer cell cox-2 passage on colon HT29 cancer cell proliferation and migration.
