摘 要:研究采用DNA池结合PCR产物直接测序及PCR-SSCP技术对贵州白山羊和贵州黑山羊GHSR基因进行单核苷酸多态性检测,并探讨其与生长性状的关系。结果发现,在这两个山羊品种GHSR基因外显子2和3’侧翼序列各检测到1个SNP(G200A和T628C)。其中,G200A为同义突变,表现为3种基因型,分别定义为GG、GA和AA,而在外显子1和5’侧翼序列未发现多态位点。χ2适合性检验表明,贵州白山羊和贵州黑山羊GHSR基因外显子2等位基因的分布符合Hardy-Weinberg平衡(P<0.05)。基因型与生长性状间关联分析显示,3个试验群体中GG基因型个体的体高显著高于GA基因型和AA基因型(P<0.05),且贵州黑山羊公羊群体中GG基因型个体的体重和体斜长均显著高于AA基因型,而其它指标间未达到显著水平(P>0.05)。初步认为:GHSR基因可望作为影响贵州地方山羊生长性状的候选基因。Currently, the DNA pool, direct sequencing of PCR products, as well as PCR - SSCP were used to detect the single nucleotide polymorphism (SNP) of GHSR gene in Guizhou white goat and Guizhou black goat. The results showed that G2OOA and T628C were detected in exon 2 and 3' UTR, however no SNPs in exonl and 5' UTR of GHSR gene were investigated in these goat breeds. The SNP (G2OOA) was a sense mu- tation and might be divided into three genotypes: GG, GA and AA. The Chi square test results indicated that the SNP frequencies of exon 2 of GHSR gene fit with Hardy - Weinberg equilibrium in this population (P 〉 0.05). Association of genotype with growth traits revealed that body height of individuals with GG genotype were higher than that of genotype GA or AA in three test groups, and the individual body weight or body length of Guizhou black goat ( 10 months) with GG genotype were higher than that of genotype AA. No signif- icant differences were obtained among the rest of traits. The available data suggested that GHSR gene might be a candidate gene involving in goat growth traits.
