摘　　要：目的：建立能测定大鼠血浆中通脉丸浓缩液中士的宁含量的UPLC分析方法。方法：大鼠血浆样品以液液萃取法处理后用BEH C18色谱柱分离,流动相乙腈-酸水（23∶77）,酸水为0.02 mol·L^-1磷酸二氢钾与0.01 mol·L^-1庚烷磺酸钠等量混合,并用10%磷酸调体系p H 2.5;流速0.18 m L·min^-1,检测波长260 nm,柱温30℃。结果：血浆中内源性物质不干扰士的宁的测定,士的宁在0.043 2~10.8 mg·L^-1线性良好,日内、日间精密度均〈10%,平均回收率均〉85%,稳定性符合体内药物分析要求。结论：该方法灵敏度高、紧密度好,可应用于通脉丸浓缩液中士的宁的药物动力学研究。Objective：To establish an effective method for determining the strychnine of Tongmai pill concentrate （TPC） in rat plasma by UPLC.Method：The rat plasma samples were processed by liquid-liquid extraction and separated on a BEH C18 column.The mobile phase consisted of 23% acetonitrile and 77% water containg 0.02 mol ·L^-1 potassium dihydrogen phosphate and 0.01 mol·L^-1 sodium heptanes sulfonate （adjusting pH 2.5 with 10% phosphonic acid.The flow rate was 0.18 mL ·min^-1 The detection wavelength was set at 260 nm and the column temperature was 30 ℃.Result：The determination of strychnine was not interferred by the endogenous in the rat plasma.The calibration curve of strychnine was in good linearity within the range of 0.043 2-10.8 mg ·L^-1.The mean extraction recoveries were more than 85% and intra-and inter-day precisions were less than 10%.The stability results meet the requirements for the biopharmaceutical analysis.Conclusion：This sensitivite and precise analytical method is applicable to the pharmacokinetic study of strychnine of TPC in rat plasmas.
