摘 要:目的研究艾灸梁门、足三里穴促进胃黏膜损伤修复的信号转导机制。方法将45只大鼠随机分为正常组(10只)、模型组(15只)、胃经穴组(10只)和对照点组(10只),除正常组外其余各组采用束缚冷应激法制作应激性胃溃疡大鼠模型,胃经穴组造模后第2天鼠板束缚艾灸"梁门""足三里"穴;对照点组选取"梁门""足三里"穴外侧旁开0.5 cm处为对照刺激点,两组均每次20 min,每日1次,持续12天。正常组和模型组仅鼠板束缚。采用Master Antibody Microarray Slides芯片检测各组大鼠胃黏膜细胞信号蛋白质磷酸化水平。结果与模型组比较,胃经穴组大鼠胃黏膜细胞10种蛋白质磷酸化水平下调,其中TRAF2、Stat1、p53、Mkp-3、NF-KB 5种蛋白磷酸化水平差异有统计学意义(P<0.01);14种蛋白质磷酸化水平上调,其中MEK、Ras、P38MAPK、PI3K、PAK、B-Raf、ERK2 7种蛋白磷酸化水平差异有统计学意义(P<0.01)。结论艾灸梁门、足三里穴可调节多种相关信号蛋白质的磷酸化水平,促进胃黏膜的损伤修复。Objective To explore the signaling mechanism of moxibustion at points Liangmen( ST21) and Zusanli( ST36) for promoting damage repair on gastric mucosa. Methods Forty-five rats were randomly divided into a normal group( 10 rats),a model group( 15 rats),a stomach meridian acupoint group( 10 rats),and a control acupoint group( 10 rats). Cold restrain stress method was applied on each group except normal group to make stress gastric ulcer rat model. On the 2nd day after modeling,rats in the stomach meridian acupoint group were astricted to rat boards and applied moxibustion at points Liangmen( ST21) and Zusanli( ST36),while the rats of control acupoint group were given moxibustion at control points which located at 0. 5 cm beside Liangmen( ST21) and Zusanli( ST36). The treatment lasted for 20 min each time,once daily for 12 days for both groups. Rats in the normal group and the model group were just astricted to rat boards. Levels of signaling protein phosphorylation in gastric mucosal cell were detected with Master Antibody Microarray Slides in each group. Results Compared with the model group,levels of 18 protein' s phosphorylation in the stomach meridian acupoint group were down-regulated,among which the decreased levels of 5 protein's phosphorylation( TRAF2,Stat1,p53,Mkp-3 and NF-KB) showed statistically differences( P〈0. 05 or P〈0. 01). Levels of twenty nine protein's phosphorylation were up-regulated,among which,the increasing levels of MEK,Ras,P38 MAPK,PI3K,PAK,B-Raf and ERK2 showed statistical significance( P〈0. 05 or P〈0. 01). Conclusion Moxibustion at Liangmen( ST21) and Zusanli( ST36) may regulate multiple related signaling protein phosphorylation levels and promote damage repair of gastric mucosa.
