佐金平木滋肺阴法对阴虚阳亢型肾性高血压大鼠血压及胸主动脉AT_1R表达的研究

摘　　要：目的:通过观测阴虚阳亢型肾性高血压大鼠(renovascular hypertension rats,RHR)血压及相关指标,探讨滋肺阴合补肝肾法对阴虚阳亢型RHR的降压机制。方法:筛选2月龄健康雄性SD大鼠78只,随机分为假手术组13只与手术组65只。制备阴虚阳亢证肾性高血压模型。筛选造模成功的阴虚阳亢证RHR,随机分为模型组、卡托普利组、滋肺阴组、补肝肾组、滋肺阴合补肝肾组(简称综合组)。光镜下观察胸主动脉AT1R表达。结果:(1)各治疗组阴虚阳亢型RHR血压的变化:1基础血压:各组均无统计学意义(P>0.05)。2手术后第二周末血压:与假手术组比较,其余各组血压均明显增高(P<0.05)。3治疗后各组血压的变化:治疗4周后,与模型组比较,卡托普利组、补肝肾组及综合组血压明显降低(P<0.05),滋肺阴组有所降低但无统计学意义(P>0.05)。(2)光镜下,AT1R阳性表达呈棕黄色颗粒,位于主动脉平滑肌细胞中。AT1R在假手术组少量表达,在模型组大量表达;与模型组比较,卡托普利组、补肝肾组和综合组AT1R阳性表达的IOD明显减少(P<0.05),滋肺阴组减少不明显(P>0.05)。结论:滋肺阴合补肝肾法(简称综合法)对阴虚阳亢型RHR有较好的降压效果,且能减少血管平滑肌细胞AT1R的表达。Objective： Through the detection of the content of the expression of AT1 R in aortic smooth muscle, investigating the hypotensive mechanism of Zorkin wood nourishing Yin and liver kidney hyperactivity renal hypertensive rats. Methods： Seventy-eight cases of two month old healthy male Sprague Dawley （SD） rats were randomly divided into the sham operation group with thirteen cases and the surgical group with sixty-five cases. The thirteen cases in the sham operation group were ligated only the left renal artery, the surgical group were treated with ligation of left renal artery to make hypertension model, two weeks later, through the tail cuff method to measure the rats＇ blood pressure, the successful model of the rat plus irrigation aconite decoc-tion of four weeks to prepare hyperactivity model. The hyperactivity hypertensive rats were randomly divided into model group, captoprill group, nourishing Yin group （Ziyin group）, took the thoracic aorta, using immunohistochemical methods to detect thoracic aortic AT1R expression, used the IPP analysis system to analysis the average value of the integral optical density of AT1 R positive expression, HE staining under the light microscope to observe the changes in the intima, Results ： （ 1 ） The change of each treatment groups towards blood pressure of hyperactivity renal hypertensive rats. ①The basis of blood pressure： the blood pressure of model group, no statistically significant （P〉0.05）. ②The blood pressure after 2weeks of the surgery the blood pressure of rats each treatment group were significantly increased （P〈0. 05）. ③Changes in blood pressure after treatment in each group.
