摘 要:目的:观察喂饲高脂饲料致"痰浊"证侯对大鼠肝脏线粒体呼吸功能损伤和三磷酸腺苷(ATP)含量的影响。方法:20只SD大鼠按随机数字表法分为普通饲料喂饲组和高脂饲料喂饲组,每组10只动物。于相同环境下分别喂饲标准配方饲料和高脂饲料30天,高脂饲料配方为:16%猪油、10%糖、6%蛋黄+基础饲料。动物模型制作完成后,乙醚麻醉,取腹主动脉血及肝脏组织,液相氧电极法检测肝细胞线粒体呼吸控制率(RCR),荧光素酶法检测肝组织ATP含量,同时测定血清肝功能指标,并行肝组织HE病理切片观察。结果:喂饲高脂饲料30天,血清胆固醇含量显著升高(P<0.01),肝细胞线粒体3态呼吸效率显著降低,RCR值降至对照组的64.88%,ATP含量降至对照组的58.96%(P<0.01);除白蛋白含量及白/球比外,血清肝功能指标较普通饲料喂饲组均显著升高(P<0.01)。病理切片显示高脂饲料喂饲组大鼠肝组织出现轻、中度脂肪变,肝小叶有少量炎性细胞浸润及散在点状坏死。结论:高脂饲料所致痰浊可造成肝细胞及线粒体呼吸功能受损,ATP合成减少,机体能量代谢功能发生障碍。Objective: To observe the Influence of high-fat diet induced phlegm syndromes on rat liver mitochondrial respiratory dysfunction and adenosine triphosphate (ATP) content. Methods: 20SD rats were randomly divided into two groups, the normal group and the high fat diet group, 10rats in each group. The normal group fed a normal diet, meanwhile, at the same environment, the high fat diet group fed high-fat forage for 30days, high-fat diet component was: 16% lard, 10% sugar, 6% egg yolk+basal diet. After successful modeling, the abdominal aorta blood and liver tissue were taken from each rat. Applyying methods of liquid oxygen electrode hepatocyte mitochondrial respiratory control ratio (RCR) and luciferase enzyme to detect liver tissue ATP levels, analysis serum to detect liver function, HE staining were observed simultaneously. Results: Compared to the normal group, after feeding high-fat diet for 30days, serum cholesterol levels of high-fat diet group were significantly increased ( P〈0. 01 ), liver mitochondrial respiratory efficiency 3-state significantly reduced, RCR value dropped to 64. 88%, ATP levels fell to 58.96% (P〈0. 01 ) ; the indication markers of liver function were higher universally except of albumin levels and albumin/globulin (P〈0. 01 ). Pathological examination showed that liver tissue of rats fed with high-fat diet had mild or moderate steatosis, a small amount of inflammatory cell infiltration and scattered punctate necrosis around lobules. Conclusion: The highfat diet-inducedphlegm can cause liver cell and mitochondrial respiratory function damaged, decreases the abilitys of ATP synthesis and disorder energy metabolism function of the liver.
