“脾肾相关”机理探讨补肾健脾方对脾肾两虚型骨质疏松症大鼠外周血中OPG、RANKL、VIP、MTL、GAS含量的影响

摘　　要：目的:部分阐明补肾健脾方治疗骨质疏松症的作用机制,从而为阐明"脾肾相关"科学内涵提供进一步的实验依据。方法:建立脾肾两虚型大鼠骨质疏松模型,并在此基础上分别给予阳性对照药、补肾方药、健脾方药及补肾健脾方药进行治疗。给药结束后,检测各组大鼠的BMD,并采用ELISA法对外周血血清中VIP、MTL、GAS、OPG、RANKL含量进行测定。结果:健脾方、补肾方和补肾健脾方均能提高脾肾两虚型骨质疏松大鼠的BMD,其中补肾健脾方的效果最佳;健脾方对MTL、GAS、VIP的调节作用强于补肾方,补肾方对OPG、RANKL的调节作用强于健脾方,而补肾健脾方对上述指标的调节作用均明显强于以上两方。结论:补肾健脾方可通过上调OPG、MTL、GAS水平,并下调RANKL、VIP水平,从而达到对脾肾两虚型骨质疏松症的治疗作用。且补肾健脾方疗效优于补肾方和健脾方,也进一步说明在骨质疏松症的发生和治疗过程中,中医的脾肾两脏是密切相关的。Objective： To partly clarify the mechanism of recuperating kidney and invigorating spleen prescription on osteoporosis, and to provide further experimental evidence for the scientific connotation of ＂spleen and kidney related＂. Methods ：Establish a rat osteoporosis model with spleen and kidney deficiency syndrome. On this basis, treat the model rats respectively with prescriptions of positive control, recuperating kidney, invigorating spleen, recuperating kidney and invigorating spleen. After the administration, measure the indicator of BMD, and the content of VIP.MTL. GAS、 OPG. RANKL in peripheral blood serum. Results： The prescription of recuperating kidney, invigorating spleen, recuperating kidney and invigorating spleen all can improve BMD of the osteoporosis rats with spleen and kidney deficiency syndrome, and the recuperating kidney and invigorating spleen prescription had the best effect. Regulatory effect of invigorating spleen prescription on MTL, GAS, VIP is better than recuperating kidney prescription, Regulatory effect of recuperating kidney prescription on OPG, RANKL is better than invigorating spleen prescription, and the Recuperating kidney and invigorating spleen prescription has the best regulatory effect on all the above indicators. Conclusion： Recuperating kidney and invigorating spleen prescription can achieve the treatment effect on osteoporosis rats with spleen and kidney deficiency syndrome by up-regulating the level of OPG, MTL, GAS, and down-regulating RANKL, VIP level. Recuperating kidney and invigorating spleen prescription was more effective than the other two prescriptions, which further demonstrates thatspleen and kidney in Traditional Chinese Medicine are closely related in the occurrence and treatment process of osteoporosis.
