摘 要:目的:探讨穿龙薯蓣皂苷对再障小鼠p-m TOR、p-S6基因及其蛋白表达的影响,解析其治疗再障的机制。方法:健康BALB/c小鼠按随机数字表法分为7组(正常组、模型组、西药对照环孢菌素组、中药对照雷公藤多苷组、穿龙薯蓣皂苷低、中、高剂量组),建立再障小鼠模型,连续给药14 d,全自动生化仪测血常规,分离培养骨髓单个核细胞,应用实时定量基因扩增荧光检测系统(QPCR)检测p-m TOR、p-S6的基因表达;应用Western Blot技术检测p-m TOR、p-S6的蛋白表达。结果:与正常对照组比较,模型组WBC、HGB、PLT计数均显著降低,治疗后各治疗组血象均不同程度上升,与模型组比较,模型组小鼠的p-m TOR、p-S6基因及蛋白表达显著上调,各组均明显高于正常组,治疗后各组均下调。穿龙薯蓣皂苷中、高剂量治疗组、Cs A组p-m TOR基因及蛋白表达与模型组比较差异有统计学意义,穿龙薯蓣皂苷组以中剂量组p-S6基因表达下调最多,疗效最好,优于中药对照雷公藤组,次于西药对照环孢菌素A组,差异有统计学意义。结论:穿龙薯蓣皂苷能下调再障小鼠p-m TOR、p-S6基因及蛋白表达,从而抑制T淋巴细胞的异常激活,发挥免疫抑制作用,改善骨髓造血功能。Objective: To study the effect of Dioscore nipponica Makino on the p-mTOR, p-S6 gene and protein expression in aplastic anemia mouse, to analysis its mechanism of treating aplastic anemia. Methods:Healthy BALB/c mice were randomly divided into 7 groups according to number table method ( normal group ; model group ; small dosage group ; middle dosage group; large dosage group; Cyclosporine A group; Tripterygium Glycoside group). After evaluating the establishment of a mouse model of aplastic anemia, these mice had gavage with different methods daily for 14 days. Blood routine examination was performed by automatic biochemical analyzer. Bone marrow mononuclear cells were isolated and cultured. Expression of p-mTOR, p-S6 gene and protein were detected by RT-QCR method and Western Blot technology respectively. Results: Compared with the normal control group, the number of WBC, HGB, PLT were significantly decreased in the model group, after the treatment, the hemogram index of each treatment group was improved in different degree, compared with the model group;p-mTOR, p-S6 geue and protein expression was significantly up-regulated in model group, each group were significantly higher than normal group. The expression of p-mTOR gene and protein in middle dosage group, large dosage group, Cyclosporine A group and model group were statistically significant,the expression of p- S6 gene declined most in middle dosage group. The effect of middle dosage is better than Tripterygium Glycoside group, worse than Cyclosporine A group. Conclusion: Dioscorea nipponica can down-regulated the expression of p-mTOR, p-S6 gene and protein in aplastic anemia mouse, then inhibit abnormal activation of T lymphocytes, suppress the immune system, improve the medullary hematopoiesis function.
